    Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells.
  • Involvement in disease

    Defects in ACTB are a cause of dystonia juvenile-onset (DYTJ) [MIM:607371]. DYTJ is a form of dystonia with juvenile onset. Dystonia is defined by the presence of sustained involuntary muscle contraction, often leading to abnormal postures. DYTJ patients manifest progressive, generalized, dopa-unresponsive dystonia, developmental malformations and sensory hearing loss.

    ab6276 staining beta Actin in SV40LT-SMC cells. The cells were fixed with 4% formaldehyde (10 min), permeabilized in 0.1% Triton X-100 for 5 minutes and then blocked in 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated with ab6276 at a working concentration of 5μg/ml and ab190573, Rabbit monoclonal [EP1332Y] to alpha Tubulin (Alexa Fluor® 647, shown in red) at 1/250 overnight at +4°C, followed by a further incubation at room temperature for 1h with an anti-mouse AlexaFluor® 488 (ab150117) at 2 μg/ml (shown in green). Nuclear DNA was labelled in blue with DAPI.
    Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

    Lanes 1 and 2: Merged signal (red and green). Green - beta actin, ab6276 observed at 42 kDa. Red - loading control, ab181602 observed at 37 kDa.

    Ab6276 was shown to specifically react with beta actin in wild-type HAP1 cells. No band was observed when beta actin knockout samples were used. Wild-type and beta actin knockout samples were subjected to SDS-PAGE. ab6276 (beta actin) and ab181602 (loading control to GAPDH) were diluted 1/5000 and 1/10 000 and incubated overnight at 4°C. Blots were developed with Goat anti-Mouse IgG H&L (IRDye® 800CW) preadsorbed (ab216772) and Goat Anti-Rabbit IgG H&L (IRDye® 680RD) preadsorbed (ab216777) secondary antibodies at 1/10 000 dilution for 1 h at room temperature before imaging.

    Western Blot of ab6276 (used as loading control) with whole tissue lysate of human grey matter from BA20 (temporal cortex).  Ab6276 was diluted 1/50000 and incubated with the sample for 16 hours at 4°C.  5 µg of lysate was loaded onto the gel, which was blocked with 5% milk for 1 hour at 20°C.  An Alexa Fluor® 680 conjugated goat anti-mouse antibody, diluted 1/5000, was used as the secondary.

    Bands below actin in image are synaptophysin (SYN).

    MDCK cells induced with increasing amounts of doxycycline to control expression of the gene of interest. All cells were normalized for loading with an albumin protein standard assay. Anti-beta actin (ab6276) was used at a concentration of 1:5000 in a milk blocking solution. B-actin blotting confirms the albumin assay in showing that an equal amount of lysate was loaded in each lane.

Afin que je puisse vous envoyer le tube de remplacement, il me faux que je trouve la commande originale. Est-ce que vous pouvez me dire soit le numéro de commande soit la date approximative de la commande, avec le nom de la personne qui avait passé la commande? Ceci est un anticorps très populaire, il m'est donc difficile à trouver la commande sans votre aide.

Aussi, vous mentionnez que vous avez optimisé la dilution de l'anticorps. Est-ce que vous vous rappelez quels dilutions vous avez testés? Effectivement, cet anticorps est un ascite et il peux donc avoir des variations de concentrations entre les lots. Nous pouvons donc pas garantir, que tous les lots vont fonctionner à 1/20000.

It appears that the information in the referenced article is incorrect. Ab26276 is thediscontinuedAnti-SorLA antibody which does not appear by name in this article. However they do identify an anti beta-actin antibody with this code in the”Pull-Down of S-Nitrosylated Proteins” section:

“β-actin (Ab-26276; Genetex Inc, San Antonio, TX)”

I did look for this at the Genetex Inc. website and did not find this product number. All the Genetex products begin with the prefix GTX-. As the reference number begins with the Abcam AB- prefix it seems likely that this group did use an Abcam product. It is also likely that they used ab6276which is ourAnti-beta Actin antibody [AC-15] and has a very similar code. We currently carry this product which has been tested in a large number of applications and species and has over 200 specific references. The link to this product is below:

I can confirm that this is the same clone. So the antibody itself is the same. The main difference between these two products are, that ab6276 is unpurified Ascite whereas ab49846 is purified. Both versions have however been tested in ICC. In order to find whether they have been tested in ICC on neuronal cells, I can recommend to look through the different publications we have for the ab6276: https://www.abcam.com/beta-Actin-antibody-AC-15-ab6276-references.html

I can also confirm, that we do guarantee all our products to work as stated on the datasheet. We do therefore guarantee ab6276 and ab49846 to work in ICC.

I have a problem with the antibody ab6276. I try to develop an indirect ELISA with this antibody, but do not get any result. This is the procedure, I've performed: - Coating of ELISA plate (Costar high bind) with actin (rabbit skeletal muscle, Cytoskeleton, inc. # AKL95)in carbonate/ bicarbonate buffer pH 9.6. Between all steps, the plate was washed 3x with TBST (200 µL wash buffer/ well) In order to find the problem, I did the following tests: Antigen adsorption study with BCA assay. Antigen did adsorb on surface with a concentration of about 1 µg/mL. Adsorption of secondary antibody on plate, detection with secondary antibody: positive, ABs react with each other. Test of secondary AB with substrate: positive. Increasing the secondary antibody concentration to 1:1000, color development was detected, but the negative control (no antigen) showed the same intensity like the samples. SDS PAGE/ Western blot with actin from the same source: positive. Do you have any suggestions? My thought is, that the protein is maybe folded in a way, that the antibody can't bind to the epitope since it is able to bind to the denatured protein in the western blot.

I would not recommend adding SDS or a reducing agent to the sample prior to coating, as this may interfere with the coating efficiency. It is possible that by boiling the protein for 5-10 minutes, the heat denaturation may be enough to expose the epitope that is being recognized by this antibody in WB.

This antibody is designed for detection of beta actin, not skeletal muscle actin. The homology between rabbit skeletal muscle actin (http://www.uniprot.org/uniprot/P68135)and the immunogen for this antibody is only 64%. Using an antibody directed against a more conserved region of actin, such as ab11003, may give better results. This antibody shows 100% homology with the above mentioned rabbit sequence.

I have a question about the epitope, ab6276 is directed against. In the data sheet is written, that the immunogen is a synthetic peptide (DDDIAALVIDNGSGL), which corresponds to the amino acids 1-15 of Xenopus laevis beta-actin. When I was looking on this amino acid sequence, I did not find the epitope, but I found EDDIAALVVDNGSGM, which is equal except 3 amino acids. But if I try to find this sequence in the human actin, I just find a 44.4% identity in 9 residues overlap 80.0% identity in 5 residues overlap 18.2% identity in 11 residues overlap 75.0% identity in 4 residues overlap and so on. My question is: Where should the epitope in human beta actin be located? Where is it meant to bind to? Or does it bind to any of these epitopes? I ordered this antibody, since it was directed against aa 1-15, and I wanted to have an antibody which is directed against an epitope at the C-terminal end for a sandwich ELISA. The capture antibody I use, is ab34731, which recognizes an epitope at the N-terminal end. If ab6276 reacts with an epitope in the middle of the protein, it might be useless for me.

The antibody ab6276 is raised against the peptide specified on the datasheet. You are correct, it does not correspond exactly to the X. laevis sequence, and we will very likely remove this note about X. laevis. Thank you for bringing this to our attention.

Reactivity with human beta actin has been demonstrated many times, though the exact location of the epitope that the binding site of the antibody recognizes on the immunogen sequence is not known. Alignment with the human sequence given at the UniProt entry at the following URL shows 87% identity (13/15):
