    Induces cartilage and bone formation. May be the osteoinductive factor responsible for the phenomenon of epithelial osteogenesis. Plays a role in calcium regulation and bone homeostasis.

    Question

    The customer has now asked:

    Are you aware whether it will or can interfere in any which way? I am going to Immunoprecipitate human chondrocytes against this antibody. I do not want any molecule of the OP-1 produced by my cells to go undetected due to some resistance issue.

    As far as I can tell the concern is that he wants to ensure that all of the OP-1 in his system will be immunoprecipitated by the antibody. Any thoughts?

    We unfortunately do not have data that would answer your customer's question about whether the antibody has more affinity for the BMP7 precursor or mature form. What is needed is a comparison of the recovery of each protein form (precursor and mature) in the IP product and in the flow-through fraction of proteins that are not precipitated.

    1) As per the data sheet there is an image showing both precursor and the mature form of human BMP7….is it specific for both forms or more for mature? Does the antibody bind both forms with equal affinity, so that the signal strength on the Western is dependent on the amount of each form in the sample?
    2) I will be using human chondrocytres isolated from cartilage for IP work. Do you have any references for cartilage?

    Answer

    The BMP7 antibody ab129156 is raised against an amino acid sequence that is present in the precursor as well as the mature protein. For western blotting, the antibody should be able to detect both forms with equal affinity, if the samples are denatured and reduced. If experimental samples contain native, non-denatured protein, the antibody may have higher affinity for one or the other, depending on their native conformation, which will determine how available the amino acid sequence is for antibody recognition. We do not have data demonstrating this, though.

    We are not aware of any references that have used this antibody for IP of cartilage, or for any assay of BMP7 in cartilage or chondrocytes.
