Specificity
The aE11 monoclonal was first reported to bind to a neoepitope exposed on polymerized C9 when incorporated into the human terminal complement complex (PubMed ID: 4035298). It was subsequently shown that this antibody also cross reacts with the C5b-8 complex and purified C8 alpha-gamma. It does not react with native C9 or C8 proteins (PubMed ID: 2424021).
ab66768 binds both membrane-bound MAC (active) and fluid-phase SC5b-9 complexes (inactive) (Mollnes TE, Harboe M (1987).

ab66768 staining C5b-9 + C5b-8 (red) in porcine liver tissue by Immunohistochemistry (Frozen sections).
Tissue was taken from allotransplants of wild-type or a1,3-galactosyltransferase gene-knockout (GTKO) pig liver, 2 hours after grafting or at necroscopy (nec). Tissue was fixed with paraformaldehyde and blocked with 20% donkey serum for 1 hour at room temperature, before being incubated with primary antibody (1/100 in diluent). -

ab66768 staining C5b-9 + C5b-8 in monkey retina cord tissue sections by Immunohistochemistry (IHC-Fr - frozen sections).
Tissue was fixed with paraformaldehyde, permeabilized with 0.5% Triton-X and blocked with 4% serum for 30 minutes at 22°C. Samples were incubated with primary antibody (1/500) for 16 hours at 4°C. An Alexa Fluor® 488-conjugated goat anti-mouse IgG polyclonal (1/300) was used as the secondary antibody. Counterstained with DAPI. Magnification: 20X.
