      Involved in degradation of small peptides. Digests preferentially peptides containing an aliphatic or hydrophobic residue in P1' position, as well as methionine, leucine or phenylalanine in P1 position of ester substrate.

      Enters the endoplasmic reticulum as an inactive zymogen and is modified by four N-linked core oligosaccharides, giving rise to a precursor known as P1 (67 kDa). As P1 transits through the Golgi, extension of its core oligosaccharides leads to the Golgi-modified P2 precursor (69 kDa). P2 is sorted away from secretory proteins at or beyond a late Golgi compartment and is subsequently delivered to the vacuole via a prevacuolar endosome-like compartment. Upon arrival in the vacuole, the N-terminal prosegment of P2 is cleaved to yield the enzymatically active mature vacuolar form of CPY (61 kDa).

      W303 background yeast strains: wild type, prc1∆ (neg control), Prc1-13xMYC (pos control) and a sec65-1 mutant.  From overnight cultures, strains were grown @30degC from OD = 0.3 to 0.6, then shifted to 38degC for 45'.  6OD pellets were collected and 0.5OD were run on SDS-PAGE (4-20% gradient).  Abcam34636 was diluted 1:5000 in 5% milk/PBS with 0.01% tween and incubated overnight 4degC.  An HRP-conj. goat anti-rabbit from SCBT was used at 1:10000 for 1 hr.  Let me know if you have any further questions.

      Question

      We used the anti-CPY antibody for WB in S. cerevisiae, which cross-reacted with all of our sample lanes (including our negative control, prc1∆, which is genetically lacking the target antigen). I can send you images of the data if you would like. For now, we are purchasing the anti-CPY [10A5B5] monoclonal antibody (ab113685). Hopefully this works better. If you could recover the cost difference between the two items, that would be great. Please let me know if you need any info (i.e. PO #) from either transaction.
