Target

  • Function

    May act as a scaffolding protein within caveolar membranes. Interacts directly with G-protein alpha subunits and can functionally regulate their activity (By similarity). Involved in the costimulatory signal essential for T-cell receptor (TCR)-mediated T-cell activation. Its binding to DPP4 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner. Recruits CTNNB1 to caveolar membranes and may regulate CTNNB1-mediated signaling through the Wnt pathway.
  • Tissue specificity

    Expressed in muscle and lung, less so in liver, brain and kidney.
  • Involvement in disease

    Defects in CAV1 are the cause of congenital generalized lipodystrophy type 3 (CGL3) [MIM:612526]; also called Berardinelli-Seip congenital lipodystrophy type 3 (BSCL3). Congenital generalized lipodystrophies are autosomal recessive disorders characterized by a near absence of adipose tissue, extreme insulin resistance, hypertriglyceridemia, hepatic steatosis and early onset of diabetes.

    Lanes 1 - 4: Merged signal (red and green). Green - ab18199 observed at 21-24 kDa. Red - loading control, ab7291 (Mouse anti-Alpha Tubulin [DM1A] observed at 55kDa.

    ab18199 was shown to react with Caveolin-1 in wild-type A431 cells in western blot. Loss of signal was observed when CAV1 knockout sample was used. Wild-type and CAV1 knockout A431 cell lysates were subjected to SDS-PAGE. Membranes were blocked in 3% milk in TBS-T (0.1% Tween®) before incubation with ab18199 and ab7291 (Mouse anti-Alpha Tubulin [DM1A] overnight at 4°C at 1 µg/ml and a 1 in 20000 dilution respectively. Blots were incubated with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) secondary antibodies at 1 in 20000 dilution for 1 hour at room temperature before imaging.

    Caveolin 1 - Caveolae Marker was immunoprecipitated using 0.5mg Mouse Heart whole tissue lysate, 5µg of Rabbit polyclonal to Caveolin 1 and 50µl of protein G magnetic beads (+). No antibody was added to the control (-).
    The antibody was incubated under agitation with Protein G beads for 10min, Mouse Heart whole tissue lysate lysate diluted in RIPA buffer was added to each sample and incubated for a further 10min under agitation.
    Proteins were eluted by addition of 40µl SDS loading buffer and incubated for 10min at 70oC; 10µl of each sample was separated on a SDS PAGE gel, transferred to a nitrocellulose membrane, blocked with 5% BSA and probed with ab18199.
    Secondary: Clean blot (HRP conjugate) at 1/1000 dilution.
    Band: 20kDa: Caveolin 1 - Caveolae Marker.

Question

I believe the reason why there was a delay was because the majority of the lab was out of the lab for a portion of the day attending a conference on campus. However, there were still people in and out of the lab all day so it might have just been a case of really unfortunate timing. Additionally, when there is only one person in the lab often times the door is shut so that could account for it as well. Besides the weekends, people will be in lab all day the coming month so it should be fine. Thanks for again for all your help and understanding, I really appreciate it. If you need anything else from me I'd be happy to provide it just don't hesitate to ask. And yes, that address is correct.

Yes, we tried all three vials. We performed them at a 1:1000 dilution, one that has worked excellent for us in the past (we have been using this great antibody from you guys for years now). We incubated the primary Cav-1 antibody at 4C overnight, and used an alexa fluor dye at 1:1000 for two hours the next day.

Having talked to my colleagues we can go ahead and replace those antibodies for you; however before we do we would like to understand why there was such a delay in the package being delivered to you, so it does not happen again.

As we talked about on the telephone yesterday, according to FedEx records they tried to deliver it twice on Monday (24th Sep) & twice on Tuesday (25th), before successfully being able to deliver it on Wednesday (26th). The reason for the failed delivery attempts was that there was nobody there to receive it. Therefore I wanted to check the shipping address with you and see if it needs to go to a different location to guarantee that someone will be there Mon-Fri, 9am-5pm. El anticuerpo Anti-Caveolin 1 (ab18199) ha sido testado en inmunoprecipitación, Western Blot e immunohistoquímica en secciones embebidas en parafina. Al haberse validado en estas aplicaciones el anticuerpo está garantizado para funcionar en tejidos de rata, y en caso de no hacerlo la garantía cubriría el reemplazo o reembolso del anticuerpo.

Como controles positivos se recomienda utilizar los siguientes lisados: NIH 3T3 (Mouse embryonic fibroblast cell line) Whole Cell Lysate MEF1 (Mouse embryonic fibroblast cell line) Whole Cell Lysate Heart (Mouse) Tissue Lysate Heart (Human) Tissue Lysate Heart (Rat) Tissue Lysate.

ab66620 es válido para trabajar en rata mediante immunohistoquímica en secciones de parafina, Western Blot e Inmunocitoquímica. ab31163 y ab18199 son validos para las aplicaciones mencionadas (IP, IHC y WB), aunque ab18199 solo lo hemos testado en immunohistoquímica en secciones de parafina, no sabemos su aplicabilidad para secciones congeladas.

En cualquier caso os recuerdo que todos nuestros productos tienen una garantía (Abpromise) de 6 meses, y en caso de no funcionar como se especifica en su datasheet, se os reemplazará el producto de forma gratuita o se reembolsará el importe del mismo.

thank you very much for your reply. I would like once more to disturb you and ask the following question; quite often you are indicating that antigen was a synthetic peptide. I am working mainly with chick brain and quite often it is impossible to understand if the antibody is reactive with chicks. I am interested if any of your caveolin antibodies (e.g. 93952, 2910 or any other) is reactive against chicken proteins and the same question with antibody 115822 (Anti-eIF2 alpha).

Here is the information I have received from our lab:

We haven’t tested this ab for crossreactivity with caveolin 2 and caveolin 3 but I’ve done a quick blast of our immunogen sequence against Mouse and it looks to be specific to caveolin 1.


If it does turn out that the KO mice are fine and it is our antibody, it would be wonderful if youcould let us know. In that case, please provide us with the experimental details and results, so that I can pass it on to our lab.
