  • Function

    Possible adhesion molecule with a role in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells. Could act as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. Presents carbohydrate ligands to selectins.

      ab64480 staining CD34 in Human kidney tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with paraformaldehyde and blocked with 10% serum for 1 hour at 20°C; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody (1/50) for 12 hours at 4°C. A Biotin-conjugated Goat anti-rabbit polyclonal (1/200) was used as the secondary antibody.

    Question

    Thanks again for helping with secondary antibody selection yesterday for an extracellular epitope for rats. I have not yet received the article you referred me to, but was thinking it over again how we can possibly prevent a non-specific binding of secondary anti –body using your 8158. Do you have any information supporting that your other CD43, your cat. # 64480 would work in ICC-IF? I would not be able to use it for cell selection, but at least can may be able to quantify hematopoietic progenitors in peripheral blood MNC.

    Answer

    That might be a better choice than ab8158, insofar as you avoid the issue of reactivity of the anti-rat IgG secondary with your sample. For instance, the anti-rabbit IgG secondary ab96889, conjugated to Dylight 488 (similar to Alexa 488) will recognize ab64480 but not IgG rat IgG in your sample. However, it has not been tested for ICC/IF (cytochemistry), only IHC (histochemistry) on paraffin-embedded tissue, so we do not guarantee it for that application. They are similar, but not identical. Further, the immunogen is from a cytoplasmic domain, so the cells will need to be fixed and permeabilized to allow the antibody access to the cytoplasm. Have you considered Thy-1 as a marker? This paper suggest it may be useful: Haematologica. 1998 Jul;83(7):587-92. http://www.ncbi.nlm.nih.gov/pubmed/9718862?dopt=AbstractPlus We have several antibodies to this target, inculding clone MRC OX-7 which we have unconjgate (ab225) and conjugated to FITC (ab226).

    Thank you for contacting us. Although we do not currently have any antibodies specifically directed against Rat CD34, we have an antibody that would be predicted to react with this protein. Ab64480 is a rabbit polyclonal antibody against human CD34. The region of the immunogen however shares 100% homology with that of Rat CD34 (SwissProt number B1PLB1). This antibody has also been characterise with IHC-P and therefore may be suitable for your customer.
