  • Function

    Identifies cytotoxic/suppressor T-cells that interact with MHC class I bearing targets. CD8 is thought to play a role in the process of T-cell mediated killing. CD8 alpha chains binds to class I MHC molecules alpha-3 domains.
  • Involvement in disease

    Defects in CD8A are a cause of familial CD8 deficiency (CD8 deficiency) [MIM:608957]. Familial CD8 deficiency is a novel autosomal recessive immunologic defect characterized by absence of CD8+ cells, leading to recurrent bacterial infections.

      ab4055 staining CD8 alpha in human spleen tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 2% BSA for 10 minutes at 21°C; antigen retrieval was by heat mediation in citric acid. Samples were incubated with the primary antibody (1/100 in TBS/BSA/azide) for 16 hours at 21°C. A biotin-conjugated goat anti-rabbit IgG polyclonal (1/300) was used as the secondary antibody.

      ab4055 at 1/500 dilution staining CD8 alpha from human PBMC (Peripheral blood mononuclear cells) tissue sections by IHC-P. The tissue was paraformaldehyde fixed and a heat mediated antigen retrieval step was performed prior to incubation with the antibody for 20 minutes. A biotinylated goat anti-rabbit antibody was used as the secondary.

    Question

    I have recently purchased a number of antibodies including ab833 (anti-Ki67, lot 68069) and ab4055 (anti-CD8, lot 71307), which I have not used with any success to date. I am currently testing these antibodies on paraffin embedded mouse sections of different tissues, but I am specifically interested in staining within the lung. I have followed the protocols described on the datasheets (I do not have access to human or mouse tonsil) and used a range of dilutions (1:25, 1:50, 1:100), incubation times (1hr RT, overnight 4C) and used microwave antigen retrieval with 10mM citrate buffer (5 and 15 mins for Ki67, and 15 and 20 mins for CD8). I have found with Ki67 that I get cytoplasmic staining rather that nuclear staining in mouse testes, and no staining in lymph nodes or spleen with CD8 but it does detect erythrocytes. I have also used both of these antibodies on frozen lung sections at a dilution of 1:100 for 90mins at RT also with no success. My protocol is outlined below. Dewax and rehydrate sections (5mins Xylene, 5mins Xylene:Ethanol 1:1, 2x 3mins 100% Ethanol, 3mins 90% EtOH, 70% EtOH, 30% EtOH, TBS+0.025% TritonX); Antigen Retrieval; 5mins 3% peroxide, 2x 3mins TBS+0.025% TritonX, 30mins 10% FCS, Primary antibody, 2x 3mins TBS+0.025% TritonX, 1hr Secondary antibody (Dako, Biotinylated Swine anti-Rabbit), 2x 3mins TBS, 45mins Streptavidin-HRP conjugate (Dako), 2x 3mins TBS, 5mins DAB, counterstain with haemotoxylin, dehydrate (reverse of above) and coverslip with Depex.

    Answer

    I'm very sorry to hear you are having problems with ab833 and ab4055. I would like to suggest using triton (0.3%v/v) in the buffers used to dilute the primary and secondary antibodies and an overnight incubation. A 10min high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining should be enough to unmask the antigen. You may wish to check if the non-specific staining you see is due to the secondary but omitting the primary antibody in a test slide. If you still have problems please get back to me and I will offer you a replacement vial as this antibody is very popular and sucessful and your vial may be damaged. Good luck with your staining,
