Target

  • Relevance

CDA (Cytidine deaminase) scavengers exogenous and endogenous cytidine and 2'-deoxycytidine for UMP synthesis. Growth inhibition of granulocyte-macrophage colony foring cells by human cytidine deaminase requires the catalytic function of the protein. It is highly expressed in granulocytes.

    Question

o echnique does not require any optimization and it works very well with all the other antibodies we have, (including abcam) we tried some of your suggestions but without any successful result.
Increasing dilution of the secondary are not decreasing the unspecifity but only increasing the time of exposure to get visible bands.
Our secondary antibody does not create any unspecific binding with any other antibody it is Affinity purified goat anti-rabbit IgG (H&L) antibody conjugated to horseradish peroxidase.
At this point we have already wasted too much time and a lot of other reagents to run blots with different conditions which have shown unsuccessful results to the antibody.
We would like to receive these two substitutions as soon as possible.

    Answer

    rdetails and for your cooperation.
I am sorry this product did not perform as stated on the datasheet and for the inconvenience this has caused.
I appreciate the time you have spent on these experiments in the laboratory and it is disappointing the results have not been successful so far. However, we often find that suggesting some scientifically thought out optimisation tips improves the results. This is obviously a preferable outcome for the customer. So I hope you can understand that our policy is to offer the best technical support possible to help optimise the results first.
The details provided in the questionnaire enable us to closely monitor the quality of our products.
As requested, I have issued a free of charge replacement
To check the status of the order please contact our Customer Service team and reference this number.
Please note that this free of charge replacement vial is also covered by our Abpromise guarantee. Should you still be experiencing difficulties, or if you have any further questions, please do not hesitate to let us know.
I wish you the best of luck with your research.

    Asked on May 17 2012

    Answer

Thank you for taking time to complete our questionnaire. I am sorry to hear that this antibody is not providing satisfactory results.
The details provided will enable us to investigate this case and will provide us with vital information for monitoring product quality.
Having reviewed this case, I would like to offer some suggestions to help optimize the results from ab38012 and ab56053. I would also appreciate if you can confirm some further details:
Overall, your protocol looks all right. However, the secondary concentration is unusual high. Have you considered a non-primary control? In addition, I would advise to try the antibody in a much lower dilution, at least 1:10000.
Another point is the specificity of the secondary antibody, which is a bit confusing: Does this antibody react against human?

Should the suggestions not improve the results, please do let me know.
In the event that a product is not functioning in the species and applications cited on the product datasheet (and the problem has been reported within 6 months of purchase), we would be pleased to provide a free of charge replacement, credit note, or refund.

    Answer

Thank you for calling us and for alerting us to the problem you are experiencing with our product. We take product complaints very seriously, and investigate every product that we feel may not be performing correctly.
As we discussed on the phone, I am our questionnaire so that we can gather further information regarding the samples tested and the protocol used. Once we receive the completed questionnaire, we will look at the protocol and see if there are any suggestions we can make that may improve the results. This information will also allow us to investigate this case internally and initiate additional testing where necessary. If the product was purchased in the last six months and is being used according to our Abpromise, we would be happy to replace or refund the antibody.
I look forward to receiving your reply.
