Target

  • Relevance

    CGRP is present in C-cells of the thyroid and in central and peripheral nerves. CGRP has several important physiologic roles: it is a potent vasodilator, and can affect the force and rate of heart beat; it can modulate acetylcholine receptor function at the neuromuscular junction; it has been demonstrated to block tolerance to morphine; and it can modulate antigen presentation in Langerhans cells in the skin.

      IHC image of CGRP staining in a Rat normal thyroid formalin fixed paraffin embedded tissue section, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab47027, 0.1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

       

      For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

      IHC-P image of CGRP staining on Rat spinal cord sections using ab47027 (1:400). The sections were deparafinized and subjected to heat mediated antigen retrieval. The sections were then blocked using 1% BSA at 21°C for 10 min. The primary antibody was used 21°C for 2 hours.
