    Storage in frost-free freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.

    Many cellular activities depend on the interaction of cells with the surrounding extracellular matrix (ECM). Most cells, in intact tissue and in culture, are attached to an ECM. Epithelial cells are associated with the basement membrane; fibroblastic cells are usually embedded in a pericellular mesh of fibrils, and tissue culture cells usually grow on a substrate which is covered by various ECM components. Studies have indicated that the matrix or its various isolated components provide not only adhesive surfaces for cells to grow on but also have effects on the rate of cell growth, mobility, morphogenesis and differentiation. Within the ECM several glycoproteins and proteoglycans have been identified. It has been proposed that the different constituents interact with each other in a rather complex fashion. The poor antigenicity of proteoglycans especially their glycoaminoglycan (GAG) moieties make it difficult to localize these molecules in tissue and cell culture. Monoclonal Anti-Chondroitin Sulfate can be used to study chondroitin sulfate proteoglycan (CSPG) distribution and its relationships to specific cell-substrate contacts.

    This antibody was developed using Goat Anti-Mouse IgM, Cy3™ conjugate.
    Immunocytochemistry analysis of cow mammary gland epithelial cells labeling Chondroitin Sulfate with ab11570 at 2.5 μg/mL. Cells were fixed and permeabilized with 4% Paraformaldehyde and 0.5% Triton X-100. The antibody was developed using Goat Anti-Mouse IgG, Cy3™ conjugate. Cells were counterstained with DAPI (blue) to stain nuclei.

wir wuerden gern verschiedene Antikoerper bestellen und haetten zu zwei Antikoerpern noch eine Frage.
1. robo3 (ab77261):
Wir wuerden den Antikoerper gern fuer Immunostainings in Zebrafisch benutzen, leider ist das bei den Applications nicht aufgefuehrt. bedeutet dies das der Antikoerper wirklich nur nicht getestet wurde, oder ist er villeicht nicht fuer diese Anwedung geeignet? Koennen Sie vielleicht eine Aussage trefen wie hoch die Wahrscheinlichkeit ist das er in Immunostainings fuer Zebrafisch ueberhaupt funktioniert, wenn Zebrafisch nur mit "predicted to work in" angegeben ist?
2. Anti-Chondroitin Sulfate antibody (ab11570)
Auch diesen Antikoerper wuerden wir gern in Zebrafisch benutzen. Leider taucht der gar nicht in der Spalte "cross reactivity" auf. auch nicht als "predicted". Koennen sie vielleicht eine Aussage treffen wie ob er uberhaupt mit Zebrafisch reagieren wuerde und wie hoch die Wahrscheinlich keit waere?

Inquiry: If the GAG portion of the Chondroitin Sulfate Proteoglycan is enzymaticaly cleaved with Chondroitinase ABC, will the CS-56 still bind to the Proteoglycan core? Will the CS-56 Ab bind to the degradation products of Chondroitinase ABC (D-glucuronic acid and N-acetyl-D-galactosamine)?

Answer

Unfortunately we have not tested this product with digested chondroitin sulphate and so it is difficult to predict the outcome. GAGs alone are poorly antigenic and so the antibody may not interact with the D-glucuronic acid and N-acetyl-D-galactosamine from the chondroitinase reaction. We have not carried out epitope mapping for this antibody and so again we cannot say for certain whether product ab11570 will bind the proteoglycan core.

Thank you for contacting us yesterday in regards to the Anti-Chondroitin Sulfate antibody [CS-56] (ab11570).

I have had a look into which tissues may serve as positive controls with the antibody. As the Chondroitin sulfate is a major component of the extracellular matrix I would expect most tissues to show positive staining. I have not been able to find any good references referring to the distribution of the target. As discussed over the phone, we have had reports of brain tissue being used:

I have also found reference to CS being abundant in fetal liver tissue with lower levels in adult liver tissue:

As there are several different chondroitin sulfate proteoglycans such as aggrecan, verican and CD44 it is difficult to determine the exact expression profile but I would expect both the muscle, skinand bone tissue you have access to show some positive staining as indicated from the following references:

The 1.7mg/ml concentration is the antibody concentration which was determined by


Radial immunodiffusion (RID) method. For your information, the ascites concentration with other proteins included is actually 38mg/ml.

Could you, pleaseconfirm if this antibody reacts with sheep cartilage?

One more question, what kind of predigestion you recommend with this antibody?

Thank you for your reply.

I can confirm that Anti-Chondroitin Sulfate antibody [CS-56], (ab11570) has been raised against the chicken protein. I am sorry this antibody has been sources externally, and regrettably we are not able to obtain the chicken sequence in order to align this with the sheep. The sheep sequence also appears to be unavailable.

For antigen retrieval,there has been an Abreview submitted by another customer who has used a commercially available antigen retrieval solution(Trilogy). I can recommend that antigen retrieval will often require some optimization.You may like to consider trying citrate bufferretrieval pH 6with several time points (3, 5, 10 and 20 minutes) to start with.
