Contains 1 fibrillar collagen NC1 domain.
    Contains 1 VWFC domain.
  • Domain

The C-terminal propeptide, also known as COLFI domain, have crucial roles in tissue growth and repair by controlling both the intracellular assembly of procollagen molecules and the extracellular assembly of collagen fibrils. It binds a calcium ion which is essential for its function.
  • Post-translational
    modifications

Proline residues at the third position of the tripeptide repeating unit (G-X-P) are hydroxylated in some or all of the chains. Proline residues at the second position of the tripeptide repeating unit (G-P-X) are hydroxylated in some of the chains.
O-linked glycan consists of a Glc-Gal disaccharide bound to the oxygen atom of a post-translationally added hydroxyl group.

Question

Thank you for your response a few weeks ago. We went ahead and purchased the antibody (ab36064) and are in the process of testing it for 2 new applications (immunoprecipitation of collagen molecules from human blood using Invitrogen’s Dynabead system and Western Blot from human serum). Per your testing policy, I would appreciate a personal discount code.

Answer

Thank you for contacting Abcam.

The testing discount codes are normally issued before you purchase the antibody, this is so we can track it in our system. However in this case I will make an exception and issue the code post purchase. To do this, could you please provide me with the Abcam order number for ab36064, so that I can generate a code.

I look forward to your reply and issuing the code.

Question

I am interested in a non-specific antibody for immunoprecipitating collagen peptides from human serum. Can you give me your impressions on whether this antibody is suitable? Do you know what the epitope(s) are, and whether the antibody binding depends on the collagen having an intact triple helix structure? Along those lines, on a western blot, would reducing vs. non-reducing conditions affect the detection?

Answer

Thank you for contacting Abcam.
I have spoken to the lab about the epitopes that are recognized by ab36064 and the epitope has not been mapped for this antibody and because it is a polyclonal antibody, it will recognize multiple epitopes.
I believe it would be suitable for your needs since it does recognize multiply collagen family members. As to whether using this antibody under reducing conditions would affect it, I cannot say for certain, but I do not think it will. Although since we have not tested this antibody in western blot, we cannot guarantee that it will work, therefore you could take advantage of our testing discount program:
