    Mitochondrial localization of COX5A. Cultured Human embryonic lung-derived fibroblasts (strain MRC5) were fixed, treated for heat-induced antigen retrieval, permeabilized and then labeled with ab110262 (10 µg/ml) followed by an AlexaFluor® 488-conjugated-goat-anti-mouse IgG2a isotype specific secondary antibody (1 µg/ml).

Thanks for sending the western blot data to my Outlook account. I agree it is very strange that the powerpoint file somehow turned into one of our datasheets!
I agree that the vial of ab110262 you received is not working as expcted. I am happy to send a vial of MS408. This corresponds to catalog number ab110261. The current lot we have in stock is GR47109-2. Please let me know if this is OK and I'll send out this replacement immediately.

Question

Please see the attached image of another experiment with a negative
result that we obtained with Ab110262 COX5a lot GR73589-1. Moreover, we don't
need to strip the membranes prior to the second blot (with CORE2), since
the two proteins have two very different molecular weights.
For this reason, could you please send us in substitution the antibody COXIV
MS408, lot B0659?

Thank you for your enquiry. I am sorry to hear this vial of ab110262 is giving you trouble.
And thanks for including the western blot images. If I understand you correctly, you first probed the membrane with anti-CORE2 antibody and then probed the same membrane with COXVA antibody. It appears that the essentially the same signal is showing through on the COXVA staining as was present on the CORE2 staining.
I think the first blot was not completely stripped of signal. So the second blot is difficult to interpret.
I would recommend running a new gel and blotting for COXVA.
I hope this is helpful and I look forward to hearing if these suggestions prove useful.
