    This gene encodes a 178 aa mature protein that is found in the mitochondrion and may participate in the permeability transition pore. While technically this protein is Cyclophilin F, literature references commonly refer to this protein as 'cyclophilin D’ or ‘CypD’. A different cytoplasmic protein of 370 aa, represented by Entrez GeneID 5481, is identified as Cyclophilin D. This antibody does not react with this 370 aa cytoplasmic protein.

    ab110324 detected the expected band for Cyclophilin F in wild type HAP1 cells and the band was not seen in Cyclophilin F knockout HAP1 cells. Wild-type and Cyclophilin F knockout samples were subjected to SDS-PAGE.  Ab110324 and ab181602 (Rabbit anti GAPDH loading control) were incubated overnight at 4°C at 1 ug/ml and 1/20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.

     ab110324 was shown to react with Cyclophilin F in wild-type HEK-293T cells in western blot. Loss of signal was observed when knockout cell line ab266077 (knockout cell lysate ab257039) was used. Wild-type HEK-293T and PPIF knockout HEK-293T cell lysates were subjected to SDS-PAGE. Membrane was blocked for 1 hour at room temperature in 0.1% TBST with 3% non-fat dried milk. ab110324 and Anti-GAPDH antibody[EPR16891] - Loading Control (ab181602) overnight at 4°C at a 1 in 10000 dilution and a 1 in 20000 dilution respectively. Blots were developed with Goat anti-Mouse IgG H&L (IRDye®800CW) preadsorbed (ab216772) and Goat Anti-Rabbit IgG H&L (IRDye®680RD) preadsorbed (ab216777) secondary antibodies at 1 in 20000 dilution for 1 hour at room temperature before imaging.

Sachez que ces anticorps sont contre la cyclophiline F ', PPIF, CYP3, SwissProt: P30405. Toutefois, dans le domaine mitochondrial, cette protéine est généralement appelé CYPD ou cyclophiline D - une protéine de 25 kDa dans les mitochondries.
Je peux vous confirmer qu’ab110324, ab64935 et ab3567 ne croisent pas avec les Cyclophilines A, B et C. Cependant, pour ab86425 et ab37664 nous n’avons pas déterminé si ces anticorps croisent avec les Cyclophilines A, B et C.

Customer odered this product as she thought it is the mitoch. expressed protein. However, there are two proteins labbeled as Cyclophilin D, and this abs detects the cytoplasmatic form. Unfortunately, it's not very clear on the ds. The same is vice versa for the mitochondrial. This info can be found, but WAY down on the ds.
Worse, the ds for the cytoplasmatic abs, ab135717 had been changed in Oct 2012.

The antibody was raised against full length recombinant rat CypD protein.

The identity between cyp A and “cypD” (cylophilin F, PPIF) is only 75% which is not high.

The antibody shows no bands in CypD knockouts reported in the literature, also blotting with mitochondrial fractions and immunofluorescent microscopy indicate only a mitochondrial location.

Therefore, we feel vert confident in the specificity of this antibody.

Hi Abcam,

A customer would like to know what the predicted band size is for this antibody.

On the image provided on the datasheet the predicted band size is 41kDa, but this looks more like 22 kDa.

Could you please confirm the above?

I have investigated the details for ab110324 Cyclophilin F antibody,I can confirm that the correct predicted molecular weight, according to the WB data on the datasheet and the SwisProt/Uniprot protein database link for Cyclophilin F is 22 kDa.

The datasheet has been updated with the correct information and I apologize for any confusion.
