Relevance
Transcriptional repressor of genes that require a bHLH protein for their transcription. Likely to be involved in the functional rather than the morphological differentiation of neurons. Loss of DPN function results in weak motor activity, lethargic behavior, and death. Implicated in sex determination as Groucho-DPN complex act directly to repress SXL transcription -

Immunohistochemistry analysis using ab195173 detecting the nuclear transcription factor Deadpan protein in the neuroblasts of the third instar larval brain of Drosophila melanogaster. The brain was fixed in 4% formaldehyde (20 min) and then incubated in 1%BSA / 10% normal goat serum in 0.1% PBS-Tween for 1hr to permeabilise the cells and block non-specific protein-protein interactions. The brain was then incubated with ab195173 (10 µg/mL) overnight at +4°C and detected with Alexa Fluor® 555 goat anti-rat IgG. The counterstain was chicken anti-GFP detected with Alexa Fluor® 488 used according to manufacturer’s instructions.
