    Component of intercellular desmosome junctions. Involved in the interaction of plaque proteins and intermediate filaments mediating cell-cell adhesion.

    Epidermis, tongue, tonsil and esophagus.

    Defects in DSG1 are the cause of palmoplantar keratoderma striate type 1 (SPPK1) [MIM:148700]; also known as keratosis palmoplantaris striata I. SPPK1 is a dermatoligical disorder characterized by thickening of the skin on the palms and soles, and longitudinal hyperkeratotic lesions on the palms, running the length of each finger.

    I spoke with antibody services this morning. he asked me to send protein sequences for desmoglein-1 and -3 so that we may find an antibody predictive of binding to these proteins in horse laminar tissue.     Answer

    Thank you for contacting us. Although we do not have DSG 1 and 3 antibodies that have been tested for reactivity with horse or even in all the applications you anticipate using, we do have at least one DSG1 antibody that I think will be useful.

    The catalogue number is ab124798:

    The immunogen sequence is 93% conserved in the horse sequence you sent (14 of 15 amino acids are identical), so it is likely that ab124798 will cross-react with the DSG1-like horse protein. The antibody has not been tested in IHC on frozen tissue sections, only paraffin-embedded tissue. It has not been tested by flow cytometry, but does work in immunocytochemistry, and is likely to work for flow cytometry as well.

    For DSG3, the best candidate is ab14416, a mouse monoclonal raised against the extracellular portion of human DSG3. This region is 84% conserved in the horse sequence you sent. We do not know where the epitope that ab14416 recognizes falls within the extracellular region, so it is impossible to say if the epitope (generally consisting of a 6 - 8 amino acid sequence) is more or less than 84% identical to the homologous horse sequence. Again, the antibody has not been tested in all the applications you specified.
