Target

  • Relevance

    FITC (fluorescein isothiocyanate) is a fluorochrome dye that absorbs ultraviolet or blue light causing molecules to become excited and emit a visible yellow-green light. This emission ceases upon removal of the light causing the excitation. Fluorochrome labeling provides rapid, accurate localization of antigen-antibody interaction when one of the reactants is part of a cell, tissue or other biological structure. FITC is a commonly used marker for antibodies in immunofluorescent techniques since the conjugation of FITC to proteins is relatively easy and does not, in general, destroy the biological activity of the labeled protein. FITC is widely used as a hapten to label different proteins.

Question

I was very concerned, but having received your message did spend several days of time and effort to test each of these antibodies. Unfortunately, none of the antibodies worked in IF. The high temperature and long transition time must have caused significant degradation. We order the vast majority of our antibodies from Abcam and have never had this problem before.

I need your assistance in arranging for a replacement of these antibodies a soon as possible. I am under considerable time pressure to complete a series of experiments involving these antibodies, which are critical for an important upcoming grant deadline. It was also a significant cost of about ˜$1,200.00

Please let me know what I can do to initiate this process. Is it possible to have the replacement antibodies shipped out to the lab? This would be a tremendous help!

Thank you for confirming these details. I am sorry to hear that these antibodies did not work in ICC/IF.
As requested, I have issued a free of charge replacement of ab3851, ab52060, and ab112511. These will be shipped out tonight for delivery tomorrow morning. It looks like that that original order was shipped on a Saturday for Monday's delivery. Thank you for contacting Abcam.

I have been in contact with the lab and while we have not tested this clone directly with acetylated FITC, the antibody was tested with carboxy-fluorescein diacetate succinimydil ester (CFSE) in immunohistochemistry to trace CFSE-labeled lymphocytes in two-color staining (anti-FITC developed by PO-conjugated anti-mouse IgG) in combination with rat monoclonal antibodies developed with alkaline phosphatase. Since the acetyl groups are hydrolyzed within the cells after the uptake followed by the amino-reactive binding via the succinimidyl moiety, we most probably detect liberated (deacetylated) fluorescein, but have no direct data (by competition ELISA or anything) on whether plain FDA (fluorescein-diacetate) would be recognized by F4/1.

The antibodies that have FITC conjugated to them do not need the anti-FITC antibody ab112511, if you simply want to detect FITC fluorescence by flow cytometry.
