    Target

    • Function

Actin-binding protein that enhances membrane ruffling and RAC activation. Enhances the actin-bundling activity of LCP1. Binds calcium. Plays a role in RAC signaling and in phagocytosis. May play a role in macrophage activation and function. Promotes the proliferation of vascular smooth muscle cells and of T-lymphocytes. Enhances lymphocyte migration. Plays a role in vascular inflammation.
    • Tissue specificity

Detected in T-lymphocytes and peripheral blood mononuclear cells.
    • Sequence similarities

      Contains 2 EF-hand domains.
    • Post-translational
      modifications

      Phosphorylated on serine residues.
    • Cellular localization

Cytoplasm > cytoskeleton. Cell projection > ruffle membrane. Associated with the actin cytoskeleton at membrane ruffles and at sites of phagocytosis.

        Flow Cytometry - FITC Anti-Iba1 antibody [1022-5] (ab15691)

Overlay histogram showing THP-1 (human monocytic leukemia cell line) cells stained with ab15691 (red line). The cells were fixed with 80% methanol (5 minutes) and then permeabilized with 0.1% PBS-Tween for 20 minutes. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab15691, 10 µl/1 x 106 cells) for 30 minutes at 22ºC. Isotype control antibody (black line) was mouse IgG2b (FITC) (ab91368, 10 µl/1 x 106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in THP-1 cells fixed with 4% paraformaldehyde (10 minutes)/permeabilized in 0.1% PBS-Tween used under the same conditions.

      Question

Thank you very much for your response.

I prefer to the conjugates for the excitation wavelength of 488nm and emission wavelength of 575nm. The conjugates you suggested might not work for my machine.

Do you have another conjugation available?
