Function
Hyperpolarization-activated ion channel exhibiting weak selectivity for potassium over sodium ions. Contributes to the native pacemaker currents in heart (If) and in neurons (Ih). Produces a large instantaneous current. Activated by cAMP. Modulated by intracellular chloride ions and pH; acidic pH shifts the activation to more negative voltages. -

Highly expressed throughout the brain. Detected at low levels in heart. -

Immunocytochemistry analysis of differentiated SH-SY5Y (human neuroblastoma cell line from bone marrow) cells labelling HCN2 with ab84817 at 1/100 dilution followed by secondary antibody (Alexa Fluor® 488) (green). Cells were counterstained with phalloidin (Alexa Fluor® 647) (red), and with Anti-beta III Tubulin antibody (Alexa Fluor® 555) (magenta). Nuclear DNA was labelled with Hoechst (blue).

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-HCN2 antibody [S71] (ab84817)
Immunohistochemistry (Bouin-fixed paraffin-embedded sections) analysis of human hippocampus tissue labelling HCN2 with ab84817 at 1/100 dilution (1 hour at room temperature). FITC Goat Anti-Mouse (green) was used as the secondary antibody at 1/50 dilution (1 hour at room temperature).

Immunohistochemistry (Frozen sections) analysis of mouse brain tissue labelling HCN2 with ab84817 at 1/1000 dilution (1 hour at room temperature). Tissue fixed with 10% Formalin Solution for 12-24 hours at room temperature. Secondary Antibody: HRP/DAB Detection System: Biotinylated Goat Anti-Mouse, Streptavidin Peroxidase, DAB Chromogen (brown) for 30 minutes at room temperature. Counterstained with Mayer hematoxylin (purple/blue) nuclear stain at 250-500 µL for 5 minutes at room temperature.
