Applications

The Abpromise guarantee

Our Abpromise guarantee covers the use of ab4270 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF
1/100 - 1/100.
Flow Cyt
Use at an assay dependent concentration. PubMed: 18250156

ab171870 - Rabbit polyclonal IgG, is suitable for use as an isotype control with this antibody.

WB (1)
Use a concentration of 1 - 2 µg/ml. Detects a band of approximately 32 kDa. Additional proteins at 66 kD and 100 kD were detected. It detected Histone H1 from acid extracts of colcemid treated HeLa cells but not untreated HeLa cells. Acid extracts from colcemid treated and untreated HeLa cell were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho- Histone H1 (2 µg/ml).

Answer

Concerningab4270, Histone H1, phosphorylated PabRabbit x:

Thisantibody is produced by using hyper-phosphorylated histone H1 as the immunogen. The number and location of phosphorylation sites that thisantibody recognizes have not been determined. Thisantibody has been used to monitor passage through mitosis and the signal is maximal at metaphase. The presence ofhyper-phosphorylated histone H1has also been used as a measure of cdck1 activity.

There is no single specific phosphorylation site on H1 that is required for recognition by ab4270.
