Target
-
Tissue specificity
Predominant hexokinase isozyme expressed in insulin-responsive tissues such as skeletal muscle. -
Pathway
Carbohydrate metabolism; hexose metabolism. -
Sequence similarities
Belongs to the hexokinase family.
Contains 2 hexokinase domains. -
Domain
The N- and C-terminal halves of this hexokinase show extensive sequence similarity to each other. The catalytic activity is associated with the C-terminus while regulatory function is associated with the N-terminus. Each domain can bind a single glucose and Gluc-6-P molecule. -
Cellular localization
Mitochondrion outer membrane. Its hydrophobic N-terminal sequence may be involved in membrane binding. - Information by UniProt

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - HRP Anti-Hexokinase II antibody [EPR20839] (ab237315)
IHC image of Hexokinase II staining in a section of formalin-fixed paraffin-embedded human pancreas adenocarcinoma*, performed on a Leica BOND™.
The section was pre-treated using heat mediated antigen retrieval with Tris/EDTA buffer (pH9, epitope retrieval solution 2) for 20mins. The section was then incubated with abab237315, 1/100 dilution, for 15 mins at room temperature. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. The inset negative control image is taken from an identical assay without primary antibody.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
