This antibody is specific to a 55 kDa protein known as interleukin 2-receptor (IL-2R alpha). IL-2R alpha is over expressed on the surface of activated T cell, HTLV-1 or HTLV-2 positive T cell lines and moderately expressed on activated B cells, NK-cells and macrophages. -

Involvement in disease
Genetic variations in IL2RA are associated with susceptibility to diabetes mellitus insulin-dependent type 10 (IDDM10) [MIM:601942]. A multifactorial disorder of glucose homeostasis that is characterized by susceptibility to ketoacidosis in the absence of insulin therapy. Clinical fetaures are polydipsia, polyphagia and polyuria which result from hyperglycemia-induced osmotic diuresis and secondary thirst. These derangements result in long-term complications that affect the eyes, kidneys, nerves, and blood vessels. -

ab9496 - immunohistochemistry
Formalin fixed paraffin embedded human tonsil stained with CD25, using ABC and AEC chromagen.

Overlay histogram showing Jurkat cells stained with ab9496 (red line). The cells were fixed with 80% methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab9496, 1/50 dilution) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.
