Function
Cytokine with immunoregulatory activity. May promote the transition between innate and adaptive immunity. Induces the production of IgG(1) and IgG(3) in B-cells (By similarity). May play a role in proliferation and maturation of natural killer (NK) cells in synergy with IL15. May regulate proliferation of mature B- and T-cells in response to activating stimuli. In synergy with IL15 and IL18 stimulates interferon gamma production in T-cells and NK cells. During T-cell mediated immune response may inhibit dendritic cells (DC) activation and maturation. -

Expressed in activated CD4-positive T-cells but not in CD8-positive T-cells, B-cells, or monocytes. -

This data was developed using ab227837, the same antibody clone in a different buffer formulation.
Flow Cytometry (Intracellular) analysis of Th17 polarized mouse splenocytes treated with 500 ng/ml PMA and 500 ng/ml ionomycin for 1 hour, followed by addition of 1 μg/ml Brefeldin A for another 4 hours. Cells were fixed with 4% paraformaldehyde, permeabilized with 90% methanol, and labeling IL-21 with ab282737 at a 1/500 dilution (Right). A Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at a 1/2000 dilution. Rabbit monoclonal IgG (ab172730) was used as an Isotype control (Left).
IL21 is mutual exclusively expressed with IFN gamma (Th1 marker).
Cells were provided by an anonymous collaborator.
