Target

  • Function

    The alpha-V integrins are receptors for vitronectin, cytotactin, fibronectin, fibrinogen, laminin, matrix metalloproteinase-2, osteopontin, osteomodulin, prothrombin, thrombospondin and vWF. They recognize the sequence R-G-D in a wide array of ligands. In case of HIV-1 infection, the interaction with extracellular viral Tat protein seems to enhance angiogenesis in Kaposi's sarcoma lesions.

      ab16821 staining Integrin alpha V in Human melanoma cells by intacellular Flow Cytometery. Samples were prepared by trypsinizing cells, treatment with STI and spun out trypsin, kept on ice during experiment duration. FSC and SSC were used to gate healthy cell population, which excluded PI positive cells. The primary antibody was diluted 1/250 and incubated with sample for 1 hour at 4°C in 2% FCS/0. 1%NaAzide/PBS. An Alexa Fuor® 488-conjugated Goat polyclonal to mouse IgG was used at dilution at 1/500 as secondary antibody.

    I have already purchased the antibody below (clone 10D5). I'm looking for a monoclonal antibody which binds to human alphaV protein. I will need it for flow cytometry, so ideally it will be a direct conjugate to a fluorochrome. I see on your website, that you se_l_l #ab16821 _Anti-Integrin alpha V antibody [272-17E6]. Is the epitope for this antibody known? Does it bind to alphaV when it is bound to the beta subunit? In my case, I would like it to bind to alphaV when it is expressed as alphaV/beta6. _ Does it cross react with mouse alphaV as well as human alphaV?

    In fact we did immunofluorescence (not WB) following the protocol below: Note: using the same secondary Ab we received very good results with your alpha-V Mab IMMUNOFLUORESCENCE PROTOCOL The cells (Normal Human Dermal Fibroblasts) previously adhering for 2 h at 37°C with 5% CO2 on serum coated glass were treated as follows: 1.       2x PBS washes 2.       20 minutes fixation with paraformaldehyde 4% 3.       2x PBS washes 4.       30 minutes saturation with BSA (bovine serum albumin) 1% at 37°C 5.       30 minutes incubation with Mouse monoclonal anti-integrin β1 antibody  (Anti-Integrin beta 1 antibody [12G10] (ab30394) in dilutions 1:50/ 1:100/and 1:500 in PBS+1% BSA at 37°C 6.       2x PBS washes 7.       30 minutes incubation with Goat anti-Mouse IgG (H+L) 488/555 antibody (Abcam) 1:400 in PBS+1%BSA at 37°C 8.       2x PBS washes 9.       Rapid H2O wash before mounting in  Mowiol The absence of any structures resembling focal adhesion contacts was observbed, apart from alpha-v Mab. We can provode pictures if it is necessary.
