Target

  • Function

    The alpha-V integrins are receptors for vitronectin, cytotactin, fibronectin, fibrinogen, laminin, matrix metalloproteinase-2, osteopontin, osteomodulin, prothrombin, thrombospondin and vWF. They recognize the sequence R-G-D in a wide array of ligands. In case of HIV-1 infection, the interaction with extracellular viral Tat protein seems to enhance angiogenesis in Kaposi's sarcoma lesions.

    Question

    I was particularly interested in ab124968 as it specifies applications in WB and flow cytometry. Ideally I would like to use it for WB, but I have not come across its use in any other publications. Therefore if it didn’t work with WB, I wanted to see how I could use it for flow cytometry. I was also a little curious as to the positioning of the alpha v and beta 6 bands on the gel shown. The beta 6 band certainly doesn't seem to fit with the size of beta six? Would it be possible to have a reference of where ab124968 was used with WB?

    Answer

    The higher MW band (˜ 150 KDa) on the blot for ab124968 is full length Integrin alpha-V, and the lower band (˜ 25 KDa) is Integrin alpha-V light chain (http://www.uniprot.org/uniprot/P06756). At this time we have not run any experiments to characterize the faint band at approximately ˜ 85-90 KDa so we cannot say for certain that it is Integrin beta 6. C6 is the only sample we have tested thus far that produces this band.
    Unfortunately, we do not know of any specific references that have used ab124968 in western blot.
