Target

  • Function

    Histone demethylase that demethylates 'Lys-4' of histone H3, thereby playing a central role in histone code. Does not demethylate histone H3 'Lys-9' or H3 'Lys-27'. Demethylates trimethylated, dimethylated and monomethylated H3 'Lys-4'. Acts as a transcriptional corepressor for FOXG1B and PAX9. Favors the proliferation of breast cancer cells by repressing tumor suppressor genes such as BRCA1 and HOXA5. In contrast, may act as a tumor suppressor for melanoma.
  • Tissue specificity

    Ubiquitously expressed, with highest levels in testis. Down-regulated in melanoma and glioblastoma. Up-regulated in breast cancer (at protein level).

    Intracellular Flow Cytometry analysis of HeLa (human epithelial cell line from cervix adenocarcinoma) cells labeling KDM5B / PLU1 / Jarid1B with ab181089 at 1/500. Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) at 1/2000 was used as the secondary antibody (red).Rabbit monoclonal IgG (ab172730) was used as the isotype control (black). Blue - Unlabeled control, cells without incubation with primary and secondary antibodies.

    Lane 1: Wild-type HAP1 whole cell lysate (20 µg)
    Lane 2: KDM5B knockout HAP1 whole cell lysate (20 µg)
    Lane 3: HeLa whole cell lysate (20 µg)

    Lanes 1 - 3: Merged signal (red and green). Green - ab181089 observed at 170 kDa. Red - loading control, ab18058, observed at 120 kDa.

    ab181089 was shown to recognize KDM5B in wild-type HAP1 cells as signal was lost at the expected MW in KDM5B knockout cells. Additional cross-reactive bands were observed in the wild-type and knockout cells. Wild-type and KDM5B knockout samples were subjected to SDS-PAGE. ab181089 and ab18058 (Mouse anti-Vinculin loading control) were incubated overnight at 4°C at 1.9 μg/ml and 1/20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.
