Target
-
Function
Binding to cells via a high affinity receptor, laminin is thought to mediate the attachment, migration and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components. -
Tissue specificity
Placenta, striated muscle, peripheral nerve, cardiac muscle, pancreas, lung, spleen, kidney, adrenal gland, skin, testis, meninges, choroid plexus, and some other regions of the brain; not in liver, thymus and bone. -
Involvement in disease
Defects in LAMA2 are the cause of merosin-deficient congenital muscular dystrophy type 1A (MDC1A) [MIM:607855]. MDC1A is characterized by difficulty walking, hypotonia, proximal weakness, hyporeflexia, and white matter hypodensity on MRI. -
Sequence similarities
Contains 17 laminin EGF-like domains.
Contains 5 laminin G-like domains.
Contains 2 laminin IV type A domains.
Contains 1 laminin N-terminal domain. -
Domain
The alpha-helical domains I and II are thought to interact with other laminin chains to form a coiled coil structure.
Domains VI, IV and G are globular. -
Cellular localization
Secreted > extracellular space > extracellular matrix > basement membrane. Major component. - Information by UniProt

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Laminin 2 alpha antibody [EPR23924-60] (ab259345)
Immunohistochemical analysis of paraffin-embedded mouse cardiac muscle tissue labeling Laminin 2 alpha with ab259345 at 1/500 (0.484 µg/ml) dilution followed by a ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). Positive staining in basal laminae of mouse cardiac muscle (PMID: 11969289). The section was incubated with ab259345 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin.
Secondary antibody only control: Secondary antibody is a ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101).
Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins.

Immunohistochemical analysis of 4% PFA-fixed, 0.2% Triton X-100 permeabilized frozen mouse cardiac muscle tissue labeling Laminin 2 alpha with ab259345 at 1/100 (4.84 µg/ml) dilution followed by ab150077 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (Green). Positive staining on mouse cardiac muscle is observed. The nuclear counterstain was DAPI (Blue).
Secondary antibody control: Secondary antibody is ab150077 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488).
Heat mediated antigen retrieval using sodium citrate buffer (10mM citrate pH 6.0 + 0.05% Tween-20).
