Target

  • Function

    Calcium-dependent lectin involved in innate immune defense. Binds mannose, fucose and N-acetylglucosamine on different microorganisms and activates the lectin complement pathway. Binds to late apoptotic cells, as well as to apoptotic blebs and to necrotic cells, but not to early apoptotic cells, facilitating their uptake by macrophages. May bind DNA.
  • Tissue specificity

    Plasma protein produced mainly in the liver.
  • Involvement in disease

    Note=There is an association between low levels of MBL2 and a defect of opsonization which results in susceptibility to frequent and chronic infections.

    Question

    The placental proteins were harvested using the lysis buffer containing 8M Urea and 2%CHAPS supplemented with protein inhibitors. Protein samples were prepared using Laemalli buffer with beta-ME and DTT, boiled for five minutes. The proteins were resolved on 10%SDS-PAGE and in reducing conditions. The membrane was blocked in 1% non fat dry milk in TBS over night at 4 degree. The primary antibody incubation was 1:30hr to 2:00 hr at room temperature. The membrane was washed for 3-4 times (10 min/wash) using TBST (Triton X-100, 0.02%). Secondary antibody was Goat polyclonal anti mouse IgG-HRP (1:1000 dilution), incubation time 1:00 hr at room temperature. The antibody signals were detected using ECL method.

We generally recommend blocking in 5% milk in TBST and incubating the primary antibody overnight at 4 degrees. You may also wish to try changing your lysis buffer to RIPA if possible.

    Thank you for your enquiry. The antibody has been tested in IHC in human liver and brain sections, but we do not have those images unfortunately. The reference Holmskov UL Collectins and collectin receptors in innate immunity. APMIS Suppl 100:1-59 (2000) has used the antibody in immunohistochemistry and should have some images with this antibody. We currently do not sell recombinant human MBL and unfortunately I do not know of a source of such a product. You may be able to find this information from the website www.biocompare.com or the supplier Sigma,

    Question

    I have ordered product ab23457 (mouse monoclonal to mannan binding lectin), and would like to label it. Unfortunately, the buffer it is supplied in contains 15mM azide, which is inhibitory to labeling by succinimydyl esters (according to molecular probes/invitrogen). I would like to use dialysis to remove the excess azide - however, I do not know the precise composition of the buffer the antibody was shipped in (which I need to formulate the buffer to dialyze against). Could you provide me with this information?
