    The antibody reacts with a Guinea pig lymphocyte subset probably analog to human CD8 (cytotoxic/suppressor) subset. CD8 comprises 2 subunits, alpha and beta and exists as either an alpha/alpha homodimer or an alpha/beta heterodimer. Sequence suggests that guinea pig CD8 is more closely related to human than rat or mouse CD8.

Question

I purchased Anti-Negative control for mouse IgG1 Ab (ab81216) to assess the effect of other blocking ab on tumor cell growth.
But, what I found was cells treated with IgG1 ab also showed retarded growth similar with blocking ab.
I am wondering if this item is not suitable for functional assay such as cell proliferation due to, for example, sodium azide or something ?

Answer

Thank you for contacting us.

We have not heard other reports of this, but it is possible the azide in the antibody, which is present at 0.1% in the stock solution, does have an effect on the tumor growth. Confirming this would require an experiment, for instance comparing the effect of medium containing the same amount of azide to the effect of the antibody control.

Alternatively, you could remove the azide by exchanging the buffer of the stock solution. The product ab102776 will accomplish this. Here is a URL for the product:
