Question

Need IgG isotype control for IHC on paraffin sections. I have 3 mouse monoclonal antibodies which I require isotype controls for - 2 mouse are type IgG1, one is IgG1,kappa. Do I need to match for the kappa as well or can I just use a IgG1? Please can you tell me whether I can just use IgG1 isotype control for all antibodies or does the IgG1, kappa require one of the other isotype controls, such as ab18443 or ab18447?

Answer

Thank you for your enquiry. According to my literature 99% of mouse IgG immunoglobulin molecules have a kappa light chain. This is reflected in the number of IgG, kappa isotype controls in our catalogue. Therefore given that your three mouse antibodies are IgG1; with the light chain unspecified in two of them and one specified as kappa I think that it is safe to assume that you can use a single isotype control relating for all three antibodies relating to heavy chain IgG1 and kappa light chain. For example; ab18443 Mouse IgG1, kappa monoclonal (MOPC-21) - isotype control or ab18447 Mouse IgG1, kappa monoclonal [MG1-45] - Isotype control.
