Question

I have found two mouse IgG2b isotype controls which I think are suitable for my IHC-P experiments on human endobronchial biopsies, but I have a question about them:
-ab18421 clone MG2b-57 - immunogen used is Trinitrophenol + KLH, and the AB is said to react with KLH.
-ab18469 clone MPC-11 - immunogen used is also Trinitrophenol + KLH. My question is if this AB reacts with KLH like clone ab18421. In this, case do they react with different epitopes?

Answer

Thank you for contacting Abcam.

We have not tested in-house reactivity of ab18469 against KLH, so I cannot say for certain whether the antibody will or will not bind to the KLH tag.

We have also not epitope mapped these antibodies, so we cannot say exactly where they bind, but I would suspect that they bind to different regions.

Question

Hi,   I purchased a pair of antibodies to detect mouse IgG2b by sandwich assays. The antibody that I have for the capture probe is ab11566 (Rat monoclonal [LO-MG2b-1]). And I have ab99640 (Rat monoclonal [LO-MG2b-2], biotin) as the detection probe. As proof of concept, I’d like to detect known isotype control samples using the pair of antibodies that I have. Now, I’m interested in ab18421 ([MG2b-57]) and ab18469 ([MPC-11]). Which one do you recommend? Are both isotype control samples recognized by the antibodies that I have? Another question I’d like to ask is what is the meaning of clone numbers like MG2b-1 and MG2b-2? Can I know the epitope information from these numbers? Thank you.

Thank you for your enquiry.


Both you capture and detection antibodies should recognize these isotype controls. However, as these products have not been tested in sandwich ELISA we do not know whether the capture antibody will recognize the same site or otherwise interfere with the detection antibody. Although we produce many in-house antibodies, we obtain other antibodies from a wide range of sources. Therefore, comparison experiments will not often have been done. As these products have not been tested in this application and they have not been tested against one anotherI cannot recommend one isotype control over the other.

Regarding the clone numbers, these are created by the originator the antibody for identification purposes. The epitopes of these products have not been mapped to our knowledge.

The MPC-11 myeloma is desribed in the following paper:

My understanding is that the MPC-11 isotype control is not in fact a hybridoma but is simply a myeloma that secretes full-length IgG2b with no known specificity.
