Function
Cytochrome c oxidase is the component of the respiratory chain that catalyzes the reduction of oxygen to water. Subunits 1-3 form the functional core of the enzyme complex. Subunit 2 transfers the electrons from cytochrome c via its binuclear copper A center to the bimetallic center of the catalytic subunit 1. -
Involvement in disease
Defects in MT-CO2 are a cause of mitochondrial complex IV deficiency (MT-C4D) [MIM:220110]; also known as cytochrome c oxidase deficiency. A disorder of the mitochondrial respiratory chain with heterogeneous clinical manifestations, ranging from isolated myopathy to severe multisystem disease affecting several tissues and organs. Features include hypertrophic cardiomyopathy, hepatomegaly and liver dysfunction, hypotonia, muscle weakness, excercise intolerance, developmental delay, delayed motor development and mental retardation. A subset of patients manifest Leigh syndrome. -

Lane 2: COXII Immunoprecipitated from 1mg Human Heart Homogenate (HHH) Extract. ab156031 anti-COXII captures the entire Cytochrome c oxidase complex in immunoprecipitation. -
Immunocytochemical analysis of 143B (Human osteosarcoma) wild type (WT) and mtDNA lacking (?0) cells (paraformaldehyde fixed (4%, 20 min) and Triton X-100 permeabilized (0.1%, 15 min)) labeling COXII with ab156031 at 1µg/ml (red) and anti-HSP60 (ab46798) at 1/1000 (green) as a counter stain. Secondary: Alexa 594 goat anti-mouse IgG at 1/1000 and Alexa 488 goat anti-rat IgG at 1/1000. ab156031 shows good reactivity with the wild type (143B-WT) cell line, but shows no signal with the mtDNA lacking (143B-?0) cell line. Cytochrome c oxidase 2 is exclusively encoded by mitochondrial DNA. The anti-HSP60 counter stain antibody identified mitochondria in both cell lines.

Immunocytochemical analysis of COS7 (monkey kidney fibroblast) cells (paraformaldehyde fixed (4%, 20 min) and Triton X-100 permeabilized (0.1%, 15 min)) labeling COXII with ab156031 at 1µg/ml (red). Secondary: Alexa 594 goat anti-mouse IgG at 1/1000. ab156031 shows good reactivity with the COS7 cell type indicating cross reactivity with monkey cytochrome c oxidase.
