Function
Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity. -
Involvement in disease
Defects in MPO are the cause of myeloperoxidase deficiency (MPD) [MIM:254600]. MPD is an autosomal recessive defect that results in disseminated candidiasis. -

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human spleen tissue sections labeling Myeloperoxidase with Purified ab134132 at 1:1000 dilution (0.2 µg/ml). Heat mediated antigen retrieval was performed using Heat mediated antigen retrieval using Bond Epitope Retrieval Solution 1 (pH 6.0) . Tissue was counterstained with Hematoxylin. LeicaDS9800 (Bond™ Polymer Refine Detection) was used secondary antibody was used at 1:0 dilution. PBS instead of the primary antibody was used as the negative control.

Flow Cytometry analysis of HeLa (Human cervix adenocarcinoma epithelial cell, Left) / HL-60 (Human acute promyelocytic leukemia promyeloblast, Right) cells labeling Myeloperoxidase with purified ab134132 at 1/10000 dilution (0.1 µg/mL) (Red). Cells were fixed with 4% Paraformaldehyde and permeabilised with 90% Methanol. A Goat anti rabbit IgG (Alexa Fluor® 488, ab150081) secondary antibody was used at 1/2000. Isotype control - Rabbit monoclonal IgG (Black). Unlabeled control - Cell without incubation with primary antibody and secondary antibody (Blue).
Negative control: HeLa.
