    Note=Amplification of the N-MYC gene is associated with a variety of human tumors, most frequently neuroblastoma, where the level of amplification appears to increase as the tumor progresses.
    Defects in MYCN are the cause of microcephaly-oculo-digito-esophageal-duodenal syndrome (MODED) [MIM:164280]; also known as oculodigitoesophagoduodenal syndrome (ODED). Microcephaly-oculo-digito-esophageal-duodenal syndrome is characterized by variable combinations of esophageal and duodenal atresias, microcephaly, learning disability and limb malformations. Cardiac and renal malformations, vertebral anomalies, and deafness have also been described.
    Defects in MYCN are the cause of microcephaly and digital abnormalities with normal intelligence (MCPHDANI) [MIM:602585].

      Overlay histogram showing SH-SY5Y cells stained with ab16898 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab16898, 0.5µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in SH-SY5Y cells fixed with 4% paraformaldehyde (10 min)/permeabilized in 0.1% PBS-Tween used under the same conditions.

    What species is your lysate from?


    For human samples, the n-myc oncoprotein has a predicted MW of 50 kDa (http://www.uniprot.org/uniprot/P04198), but runs in WB at around 64 kDa. Cleavage products would run at smaller MWs.

This antibody has been characterised with others raised against NMYC in the following paper which may be of help to you: Identification and characterisation of the NMYC gene product in human neuroblastoma cells by monoclonal antibodies with defined specificities. Ikegaki N et al. Prodedings of National Acadamy of Science USA. 83 p5929. This indiates that epitope for this antibody lies in the second exon of NMYC and it will recognise the cleavage products from NMYC. I hope this helps.     Thank you for your time yesterday discussing the antibody problem . The researcher informed us that the Abcam antibody 16898 was diluted as recommended with PBS, and was stored at 4 degrees. After about two weeks a yellow deposit has appeared and the fluid is cloudy. According to the user, the instructions that came with the antibody clearly said not to add preservatives. This is the first antibody they use that got contaminated.

    Thank you for your researcher's further information. The information that accompanies this antibody is that the buffer is comprised of no preservatives as detailed below: Preservative: None Constituents: 50% Glycerol, 0.05M Sodium phosphate. Furthermore we recommend that the antibody is stored at -20°C and that freeze / thaw cycles are avoided. Again as stated below: Storage instructions: Aliquot and store at -20°C. Avoid freeze / thaw cycles. We DO recommend that the antibody is diluted in PBS for certain applications prior to use. However, this certainly would NOT be recommended if the intention is to store the antibody in the fridge for two weeks, especially given the lack of preservative. It is therefore of no surprise to me that the antibody has developed mould ("contaminated"). I hope this information helps, please do not hesitate to contact us if you need any more advice or information.
