  • Function

    Tetrodotoxin-resistant channel that mediates the voltage-dependent sodium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane, the protein forms a sodium-selective channel through which sodium ions may pass in accordance with their electrochemical gradient. Plays a role in neuropathic pain mechanisms.

    Ubiquitinated by NEDD4L; which promotes its endocytosis.
    Phosphorylation at Ser-1451 by PKC in a highly conserved cytoplasmic loop slows inactivation of the sodium channel and reduces peak sodium currents.

    Cell membrane. It can be translocated to the cell membrane through association with S100A10.

    Overlay histogram showing SH-SY5Y cells stained with ab93616 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab93616, 0.5µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

Answer

The specificity of these antibodies to their target Nav channel was determined by performing western blot analysis on lysates from various HEK cell lines expressing one Nav channel. These antibodies were used to detect the presence of the channel in the lysates, and it was determined that ab85015 only detects Nav1.7 and ab93616 only detects Nav1.8.
Both these products were tested in the neuroblastoma cell line SH-SY5Y due to expression in dorsal root ganglion. These products are both tested and guaranteed to be specific for their respective Nav channels in the applications and species listed on their datasheets.

Thank you for contacting us in regards to the anti-Nav1.8 antibodies ab66743, ab83936, ab93616. I am sorry for the delay in my reply, I have been seeking information in regards to the IHC protocols used with these antibodies.

The antibodies you were most interested in I believe were ab66743 and ab93616 which are both directed towards the C-terminal portion of Nav1.8. I have unfortunately found that ab66743 has not been tested in IHC, only ICC of Spermatozoa. This information information is now being updated. I am sorry that this was not made clear previously.

With the antibody ab93616, the antibody was used to stain mouse skin tissue. This was performed on PFA fixed paraffin embedded tissue. Citrate buffer (pH 6) was used for antigen retrieval using a microwave to heat on high power for 2x5minutes. The primary antibody was incubated with the slide at room temperature for 1 houra dilution of1 ug/mLin PBST. The secondary antibody was incubated for1 hour at room temperature. The results obtained can be observed in the attachment to this email.

The ab83936, raised against the N-terminal portion of Nav1.8 has also been used in IHC. Citrate buffer was again used for the antigen retrieval (pH6) using the microwave for 10 minutesThe antibody was used in a dilution of 1/300 at room temperature for 1 hour.

I would like to reassure you that both ab93616 and ab83936 would be covered by the Abrpomise and if you were to have any problems in obtaining the expected staining and it has been used as according to the datasheet you would be entitled to a replacement antibody of your choice, credit note or refund.
