Relevance
Orexin A (hypocretin 1) a 33 amino acid peptide and orexin B (hypocretin 2), a 28 amino acid peptide, are both derived from a common 130 amino acid precursor, prepro orexin. Orexin A and Orexin B stimulate food consumption when administered intracerebroventricularly to rats. Orexin gene expression in the brain is highly restricted to distinct populations of neurons located in specific hypothalamic regions, including the lateral hypothalamic area (LHA), a region implicated in feeding behaviour. Orexin A and orexin B bind to and activate two closely related G protein coupled receptors (GPCRs), termed Orexin 1 (OX1) and Orexin 2 (OX2) receptors. Dysfunction of the orexin peptide system has been linked to narcolepsy. -

Immunohistochemical analysis of paraffin-embedded rat hypothalamus tissue labeling Orexin A with ab255294 at 1/1000 dilution, followed by Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). Cytoplasmic staining in rat hypothalamus (PMID: 9419374) is observed. Counterstained with hematoxylin.
Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Rabbit specific IHC polymer detection kit HRP/DAB (ab209101).
Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution 2) for 20 mins.
The section was incubated with ab255294 for 15 mins at room temperature.
The immunostaining was performed on a Leica Biosystems BOND® RX instrument.

Direct ELISA antigen dose-response curve using ab255294 at 1 µg/mL. Antigen (mouse Orexin A peptide) concentration of 0.488-500 ng/mL. ab97048 Goat Anti-Rabbit IgG H&L (Alkaline Phosphatase) at 1/5000 was used as the secondary antibody.
