    1 ug per mL of recombinant protein ab157838 and ab157841 were immobilised in PBS on an ELISA plate overnight. After blocking in 5% BSA, primary antibody (ab119952) was added in a concentration range of 0.017-1000 ng per mL.

Question

I am planning to order 3 antibodies (ab10214, ab15, ab119952) from your catalogous to perform flow cytometry with.
These antibodies are made in mouse but I work with murine embryonic stem cells. The data sheet of the 3 antibodies are showing reactivity with mouse.
Can I use it for my murine cells? How can I avoid aspecific binding? ,

Answer

I can confirm that these three antibodies recognize the mouse protein and the host species in which they were raised is also mouse. This may cause some non-specificity signal problem mainly in immunohistochemical using tissue sections (mouse-on-mouse) due to endogenous mouse immunoglobins. In order to be able to reduce the risk of any high background signal, you may wish to select Fab fragment secondary antibodies - rather than full molecules.

In the data sheet belonging to the antibody, you show FACS analysis of HeLa cells. Would it be possible for you to send the information regarding the fixation and permeabilization steps you used in order to produce these nice histograms.

Answer

The histogram showed in the datasheet was obtained using 4% paraformaldehyde as fixative agent, and 0.1% Triton-X100 and 0.5% BSA in 1X PBS to permeabilise.
