The acidic cytokeratins have molecular weights of 56.5, 55, 51, 50, 50, 48 46, 45, and 40 kDa. The basic cytokeratins have molecular weights of 65-67, 64, 59, 58, 56 and 52 kDa.

Target

  • Relevance

    Cytokeratins, a group comprising at least 29 different proteins, are characteristic of epithelial and trichocytic cells. Cytokeratins 1, 4, 5, 6, and 8 are members of the type II neutral to basic subfamily. Monoclonal anti cytokeratins are specific markers of epithelial cell differentiation and have been widely used as tools in tumor identification and classification. Monoclonal Anti Pan Cytokeratin is a broadly reactive reagent, which recognizes epitopes present in most human epithelial tissues. It facilitates typing of normal, metaplastic and neoplastic cells. Synergy between the various components results in staining amplification. This enables identification of cells, which would otherwise be stained only marginally. The mixture may aid in the discrimination of carcinomas and nonepithelial tumors such as sarcomas, lymphomas and neural tumors. It is also useful in detecting micrometastases in lymph nodes, bone marrow and other tissues and for determining the origin of poorly differentiated tumors. There are two types of cytokeratins the acidic type I cytokeratins and the basic or neutral type II cytokeratins. Cytokeratins are usually found in pairs comprising a type I cytokeratin and a type II cytokeratin. Usually the type II cytokeratins are 8kD larger than their type I counterparts.

    I can confirm that the Anti-pan Cytokeratin antibody [AE1+AE3], prediluted (ab961) can be used for applications without further dilutions.

    However, every experiment is different and may need optimisation by the user. Some customers seem to get excellent results by diluting this prediluted antibody further. We would recommend to optimise the experiment by trying the undiluted antibody and if possible dilutions found in the literature.

    L'anticorps prédilué ab82612 (Anti-Cytokeratin [MNF116]) a une concentration entre 1 et 3 µg/ml.
    L'anticorps ab28028 (Anti-Vimentin [Vim 3B4]) est un supernageant, la concentration en anticorps ne peut donc pas être déterminée précisement mais elle est estimée entre 1 et 3 mg/ml.
    Malheureusement le laboratoire qui produit l'anticorps ab961 (Anti-pan Cytokeratin antibody [AE1+AE3]) ne désire pas communiquer la concentration de ce produit.     The antibodies ab6401 and ab11214 are from the same clone [PCK-26], so they are the same antibody with similar binding characteristics. The difference is that ab6401 is not purified and is not conjugated while ab11214 is purified and conjugated to FITC.

    The mouse monoclonal [AE1+AE3] to pan Cytokeratin, prediluted, ab961 is already diluted and ready to use for cell and tissue staining right out of the vial. You receive 6 ml of this pre-diluted antibody.

    Thank you for your enquiry and patience. I was able to obtain the following information from the originators of these antibodies: For ab11213, "this product should recognise cytokeratins 1, 4, 5, 6, 8,10, 13, 18 and 19. Based on literature, respiratory epithelia expressses cytokeratins 5, 6, 7, and 8. Thus I would believe this antibody would prove suitable but I would not have any references available specific for the use of this antibody for rat respiratory epithelia." For ab8794, "we have tested this antibody on skin, colon, pancreas and several other epithelial tissues. We have not tested in respiratory epithelia in rat, although this antibody has broad species reactivity, reacts with mouse, rat bovine, others not tested." I haven't been able to obtain any additional information regarding ab961. If I do, I will pass it on to you. This antibody is a combination of 2 clones and recognizes acidic and basic subfamilies of cytokeratins. The cocktail of these two antibodies can be used to detect most human epithelia. The acidic cytokeratins have molecular weights of 56.5, 55, 51, 50, 50, 48 46, 45, and 40 kDa. The basic cytokeratins have molecular weights of 65-67, 64, 59, 58, 56 and 52.
