Target

  • Function

    Activates insulin, somatostatin, glucokinase, islet amyloid polypeptide and glucose transporter type 2 gene transcription. Particularly involved in glucose-dependent regulation of insulin gene transcription. Binds preferentially the DNA motif 5'-[CT]TAAT[TG]-3'. During development, specifies the early pancreatic epithelium, permitting its proliferation, branching and subsequent differentiation. At adult stage, required for maintaining the hormone-producing phenotype of the beta-cell.
  • Tissue specificity

    Duodenum and pancreas (Langerhans islet beta cells and small subsets of endocrine non-beta-cells, at low levels in acinar cells).
  • Involvement in disease

    Defects in PDX1 are a cause of pancreatic agenesis (PAC) [MIM:260370]. This autosomal recessive disorder is characterized by absence or hypoplasia of pancreas, leading to early-onset insulin-dependent diabetes mellitus. This was found in a frameshift mutation that produces a truncated protein and results in a second initiation that produces a second protein that act as a dominant negative mutant.
    Defects in PDX1 are a cause of non-insulin-dependent diabetes mellitus (NIDDM) [MIM:125853]; also known as diabetes mellitus type 2. NIDDM is characterized by an autosomal dominant mode of inheritance, onset during adulthood and insulin resistance.
    Defects in PDX1 are the cause of maturity-onset diabetes of the young type 4 (MODY4) [MIM:606392]; also symbolized MODY-4. MODY is a form of diabetes that is characterized by an autosomal dominant mode of inheritance, onset in childhood or early adulthood (usually before 25 years of age), a primary defect in insulin secretion and frequent insulin-independence at the beginning of the disease.

  • Domain

    The Antp-type hexapeptide mediates heterodimerization with PBX on a regulatory element of the somatostatin promoter.
    The homeodomain, which contains the nuclear localization signal, not only mediates DNA-binding, but also acts as a protein-protein interaction domain for TCF3(E47), NEUROD1 and HMG-I(Y).

      Immunohistochemical analysis of murine pancreatic islets, staining PDX1 (red) with ab47267.

      Tissue was fixed in formalin, permeablized with 0.25% Triton X-100, and blocked in blocking solution for 1 hour at room temperature; antigen retrieval was by heat mediation in a citrate buffer. Samples were incubated with primary antibody overnight at 4°C and a fluorescent-conjugated anti-rabbit IgG was used as the secondary antibody.

    Inquiry: I am trying to characterise the expression of pdx-1 in beta cells of rat pancreas processed in formalin and embedded in paraffin wax. After antigen retrieval and permeabilisation in TBST buffer, I incubated sections with primary antibody ab47267 (1:500 in diluent from Dako) incubated at RT for 1hr and ab6112 as the recommended secondary antibody (30min at RT). What is the recommended dilution for ab6112? I am not seeing any visible staining at all after incubation with DAB and was wondering whether I need to incubate my primary antibody for longer. Is there a protocol that I could follow? Thanks

    I would like to reassure you that this antibody is tested and covered by our 6 month guarantee for IHC-P and rat samples.     Day I



    Dewax and rehydrate as normal.
    Antigen Retrieval: Heat slides in 90 ml 10mM sodium citrate for 9 min. at 30% power in a 600 watt microwave. (Solution should reach 90-95O C but not boil.) Cool to room temp,. ˜30 min.
    Wash in PBS, 3 x 2 min.
    Quench endogenous peroxidase activity in 3%H2O2 for 20 min.
    Wash in PBS, 3 x 2 min.
    Drop on 5% normal goat serum to cover sample. Inc. 1h, RT. Use 50 ml/ml serum in PBS + 1% BSA.
    Dilute a-pdx1 antibody 1:60,000 in PBS + 1%BSA. Shake off blocking solution and drop on antibody to cover sample. Inc. overnight, 4O C in humidity chamber.



    Day II



    Shake off the antibody solution and wash slides in PBS, 3 x 2 min.
    Drop on biotinylated goat a-rabbit antibody (Diluted 1:1000 in PBS + 1%BSA) to cover samples. Incubate 1 h, RT.
    After 2O Ab has been on samples for 30 min., make up ABC solution: Add 20 ml/ml reagent A + 20 ml/ml reagent B in PBS, and mix well (it has to sit for 30 min.).
    After 1 h incubation, wash slides in PBS, 3 x 2 min.
    Drop on ABC mixture to cover each sample, and incubate for 30 min. RT.
    Wash slides in PBS, 3 x 2 min.
    Drop on DAB mixture to cover samples, 1-5 min. at room temp. (DAB = 1 drop each of A, B, and C/ml dH2O; from Zymed Labs). Watch reaction on dissecting microscope until desired brown is attained.
    Wash in dH2O, 3 x 1 min.
    Counterstain with hematoxylin (or methyl green) 30 sec. – 1 min.
    Wash in dH2O for 30 seconds (or in isopropanol if methyl green used).
    Dehydrate as usual.
