Target

  • Relevance

    Albumin is a soluble, monomeric protein that makes up about half of the blood serum protein. It is a carrier protein for steroids, fatty acids, and thyroid hormones and also plays a role in stabilizing extracellular fluid volume. Albumin is synthesized in the liver as preproalbumin which has an N-terminal peptide that is removed before the nascent protein is released from the rough endoplasmic reticulum. The product, proalbumin, is in turn cleaved in the Golgi vesicles to produce the secreted albumin.

    Question

    To whom it may concern: Hello, I purchased Rabbit polyclonal to pig Albumin antibody (ab79960) to immunostain porcine periodontal ligament epithelial cells which were co-cultured with mouse fibroblast cells to distinguish one cell from the other. However, I couldn't see anything under fluorescent microscope. This is the actual protocol I've used: A) Cell fixation 1. PBS (pre-warmed) wash 2x5min each 2. 4% formaldehyde fixation for 20mins at room tempt 3. PBS wash 3x10min each B) Immunostaining 1. Permeabilize cells using 0.05% Trition X – 100 in PBS for 5mins at room tempt 2. Quick wash with PBS x2 3. Blocking for 1hr in 10% goat serum + 3% BSA blocking solution 4. Primary antibody 1:100 in blocking solution at 4C overnight 5. PBS wash 6x5min each 6. Secondary antibody 1:200 in blocking solution at room tempt for 1hr 7. Quick PBS wash x2 8. Counter stain with 0.5ug/mL DAPI (diluted in dH20) for 20~30mins 9. PBS wash 5x2min each 10. Mount the samples with fluoromount G I used Alexa fluor goat anti- rabbit 546 as a secondary antibody.     Thank you for contacting Abcam. The anti-Pig Albumin antibody has not previously been tested in ICC/IF and so we have no standard protocol for this particular antibody, but I have looked over the protocol you sent and have a few questions/tips that may help. 1 - Have you tried staining a pure culture of porcine periodontal ligament epithelial cells and not just a mixed culture? I ask in case there is a possibility that your cells of interest are not surviving in the culture. 2 - Incubating the primary and secondary antibody in PBS rather than the blocking solution, can help increase staining. 3 - You are fixing for 20 minutes in 4% PFA, have you tried fixing for less time, maybe 10-12 minutes. Over fixation of a sample can prevent the antibody from binding to its target. 4 - Using a different blocking solution, either 10% BSA or horse serum etc, could help. Hopefully some of the above suggestions will help resolve the no staining problem. If there is anything  else I can do, please do not hesitate to contact me.
