        Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Prostate Specific Antigen antibody [8301] (ab403)
        Human normal prostate. Staining is localised as cytoplasmic and secreted. Left panel: with primary antibody at 1 ug/ml. Right panel: isotype control. Sections were stained using an automated system, at room temperature: sections were rehydrated and antigen retrieved with buffers (citrate pH 6.1) in a PT Link. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 mins. They were then blocked with protein block for 10 minutes (containing casein 0.25% in PBS) then incubated with primary antibody for 20 min and detected for 30 minutes. Colorimetric detection was completed with Diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that for manual staining we recommend to optimize the primary antibody concentration and incubation time (overnight incubation), and amplification may be required.

      Thank you for your email. I can't tell you the name of the originator of ab9537 as that information is proprietary. I double-checked ab403 and ab2218, and both of these do not contain BSA or other proteins in the storage buffer. Ab403 consists of preservative: 0.1% Sodium Azide, and constituents: sodium chloride, citrate / phosphate. pH 6.0. Ab2218 consists of preservative: 15mM sodium azide, and constituents: PBS. pH 7.4.       Thank you for your enquiry and your patience. The preparation for the primary immunization was PSA/ACT-complex as in most cases the source for PSA is seminal fluid. Free PSA is mostly deriving from tumor tissue. The secondary (booster) immunization was peformed using highly purified PSA. The affinity value was determined using a standard RIA method. Cross-reactivity with PSA/ACT-complex was determined using RIA and a Scatchard's plot. Cross-reactivity of 59% means that the binding level is 59% compared to the binding to the PSA (that was used to determine the K-value). It should be kept in mind that cross-reactivities are very much dependent on the preparation, but also on the method. Hence they should not be taken as "absolute" values.       To our knowledge, none of our anti PSA antibodies have been tested in cytofluorimetry. However, ab403 is the most popular of all our anti PSA antibodies and works in many other applications so I'd recommend that you try this one.
