Use at an assay dependent concentration.

ab19483 can work in direct ELISA, but not indirect ELISA (because any secondary antibody would bind to protein A directly as well as the primary antibody, thus leading to high background – due to the nature of protein A). ab18596 as detection antibody for sELISA.

WB
1/1000 - 1/10000. Detects a band of approximately 50 kDa (predicted molecular weight: 56 kDa).

50 kDa band is due to cleavage of the signal peptide and propeptide

Target

  • Relevance

    Protein A is a surface protein of S.aureus which binds IgG molecules by their Fc region. In serum, the bacteria will bind IgG molecules in the wrong orientation on their surface which hinders opsonization and phagocytosis. Mutants of S. aureus lacking protein A are more efficiently phagocytosed in vitro, and mutants in infection models have diminished virulence. Due to its affinity for the Fc region of many mammalian immunoglobulins, protein A is considered a universal reagent in biochemistry and immunology.

    Yes, you are right. The number after the dash signifies when the lot was aliquoted. If the number directly after the "GR" is the same then it is the same lot.

    For ab19483, chicken polyclonal to Protein A, we have 16 vials of lot GR58389-4 in our US office ready to ship and an additional 30 units of lots GR58389-3 and GR58389-4 in our UK office.

    For ab18596, chicken polyclonal to Protein A (HRP), we have 1 vial of GR74278-4 in our UK office which would take a few extra days to ship to you. We have two new lots in for that antibody, but only 1 vial left of the lot you requested.

    We currently have x vials of ab19483, lot GR58389 worldwide (x in UK office, x in US office, x in Japan office.)

    For ab18596, we only have 1 vial of lot GR74278 in stock, however we do hold 8 other vials of different stocks of this antibody in the UK.

    The way we number our lots is indeed the way you mentioned. The first part of the lot number e.g GR12345 corresponds to one particular lot. The last number e.g GR12345-X corresponds to the a particular date where we receieved the antibody from our labs. This means that lots GR12345-1 and GR12345-2 of a particularly antibody originate from the same stock but were delivered to our stock rooms at different days therefore these should be identical.

    Currently we are working with two of your antibodies: Chicken polyclonal to Protein A (ab19483) and Anti-Protein A antibody-HRP (ab18596). On your product sheet I found that the Chicken polyclonal to Protein A is IgY an the Anti-Protein A antibody-HRP is IgG. However, both anti-bodies are derived from chicken, so I was thinking they should both be IgY?

    Looking on the internet I found that chicken IgG and chicken IgY are two different names for the same antibody, which are used interchangeably. However, for me it was a bit confusing. Therefore, I was wondering whether you can confirm that both these anti-bodies (ab19483 and ab18596) are the same isotype?

    Are they actually the same anti-body with and without HRP-conjugate or is there a reason why one of them is called IgG and the other IgY?

    The conjugated antibodies ab18596 and ab18598 are prepared from the unconjugated ab19483. The formulations differ (for instance, there is no azide in the HRP conjugate buffer) but the antibody is the same for all three.

    I fixed a mistake on the ab19483 datasheet regarding the purity, which was inconsistent with the other two datasheets. It is immunogen-affinity-purified.
