On the data sheet of the isotype : ab 125938 they mencioned to do antigen retrievel ( AR) with EDTA.

Does that mean that we can’t use this isotype with an antibody who needs AR with citrate ?

When we dilute te isotype 1/100 as they say on the datasheet we have a lot of background !

Please note that the Isotype control should be used along with primary antibodies and the concentration of use should match the primary antibody.

The recommended 1/100 dilution is for guidelines only, customer have to optimize the conditions themselves. Regarding the antigen retrieval, it means, this can only be used with antibodies that requires antigen retrieval step.

The isotype control should not give you any staining so if it is showing high background then protocol optimization might be needed e.g. more diluted antibody or optimization of antigen retrieval solution.

I can confirm that ab51074 antibody is sold as tissue culture supernatant. Unpurified antibodies, such as those sold as whole antiserum, ascites or tissue culture supernatant will not have a concentration stated on the datasheet. Antibody concentration is usually determined by protein assay, and serum / ascites / tissue culture supernatant will contain a lot of other proteins, which means the antibody quantification would not be accurate.

I can confirm that for tissue culture supernatant, concentration of antibody is known to very between 1 - 3 mg/ml
