Answer

Thank you for contacting us again.
For price and shipping please contact our distributor Sapphire via our website.
There are two types of light chain in humans (as in other mammals),
kappa (κ) chain, encoded by the immunoglobulin kappa locus (IGK@) on chromosome 2 and lambda (λ) chain, encoded by the immunoglobulin lambda locus (IGL@) on chromosome 22

Antibodies are produced by B lymphocytes, each expressing only one class of light chain. Once set, light chain class remains fixed for the life of the B lymphocyte. In a healthy individual, the total kappa to lambda ratio is roughly 2:1 in serum (measuring intact whole antibodies) or 1:1.5 if measuring free light chains, with a highly divergent ratio indicative of neoplasm.

Question

I'm using primary antibody F4/80 monoclonal [CI:A3-1), Cat#AB6640 (from ABCAM). I want to know if the isotype below is the most suitable one. The application is IHC-Paraffin

Rat IgG2b, kappa monoclonal [RTK4530] - Isotype control (Cat#ab18541).

Please advise me another isotype if the above is not the right one. Thanks

Question

I am looking for a rat IgG2b isotype control for ChIP experiments. I saw on your webpage that you have two antibodies (ab18536 and ab18541) that are suitable for IP. Did you test whether they also work for ChIP? If not, would it be possible to obtain a small aliquot for a test experiment? Another question is, are they really specific for IgG2b? Many thanks for your help!

Answer

Thank you for contacting us.

Unfortunately ab18536 and ab18541 have not been tested in ChIP. It doesn’t mean they won’t work in this application, but we don’t have data to support it.

If you are interested in participating in our antibodies evaluation system (Abreview), and test any of these antibodies with ChIP I will be happy to offer a discount code for a future purchase if you submit the results (in a form of an Abreview) to our website.

Therefore, I can offer a discount off a future purchase if you buy an antibody and test it in the new application and submit feedback to us in the form of an Abreview. It doesn’t matter whether the Abreview is positive or negative, we would just really like to receive your feedback.

If you are interested in this offer, please follow these steps:

1. Reply to this e-mail to let me know that you would like to proceed and test either (or both) of the antibodies in ChIP. I will then send a discount code. This code must be issued before purchasing the antibody so please wait for my reply before ordering.

2. Purchase the antibody either by phone, fax, or online (www.abcam.com).

3. Test it in the new application.

4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!).

To find out more about our Abreview system, please visit: https://www.abcam.com/abreviews.

5. After the review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. The discount can be redeemed for any primary antibody ordered and the discount code is valid for 4 months after issue.

Please let me know if you have any questions about this offer and I would be happy to help you further.

The Terms and Conditions of this offer can be found at: https://www.abcam.com/abtrial.

Please notice these are isotype control antibodies, they will not bind Rat IgG2b, but they will confirm the primary antibody is binding to the specific target, and not randomly performing non specific Fc receptor binding.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Question

I might be interested in purchasing the anti-IL-17RB monoclonal
(ab86488).
The extracellular domain of the IL17Rb receptor is said to have been
used for generating the antibody, however only western blot is listed
for its application.
I was wondering whether it has been tested for flow cytometry and it
doesn't work so it is not listed or whether it hasn't been
tested for this application yet?

Answer

Thank you for contacting us.

Flow cytometry has not yet been tested with this antibody. Therefore, we do not have data to show if and how well it would work in this application. But for UNTESTED species and/or applications, we have established a testing discount program. Here is a brief description of how it works:

The testing discount program is for customers who like to use an antibody/protein on an untested species/application. You would purchase the antibody at full price, test it and submit an Abreview with your data (positive or negative). On your next order you will receive a discount for ONE antibody at the full price (100%) of the antibody you have tested. The terms and conditions applicable to this offer can be found here: https://www.abcam.com/collaborationdiscount.

As isotype control, I would suggest either

ab18450 (rat IgG2a) or ab18541 (rat IgG2b)
https://www.abcam.com/Rat-IgG2a-kappa-monoclonal-aRTK2758-ab18450.html
https://www.abcam.com/Rat-IgG2b-kappa-monoclonal-RTK4530-Isotype-control-ab18541.html

I will check with the lab if they have more information about the specific subisotype of this antibody (2a or 2b).

Please let me know if I shall issue a testing discount code for you.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Answer

Ab18541 is an isotype control i.e a Rat IgG2b, kappa monoclonal antibody, not an antibody against a specific protein which would be expressed in some tissues. Can I please check the catalogue number you are enquiring as this product does not have positive controls?
