    IHC image of RUNX1 / AML1 + RUNX3 + RUNX2 staining in a section of frozen normal human tonsil performed on a Leica BONDTM system using the standard protocol. The section was fixed in 10% paraformaldehyde (10 min) prior to staining. The section was incubated with ab92336, 1/500 dilution, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. The inset secondary-only control image is taken from an identical assay without primary antibody.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

J'ai fait donc aussi l'alignement pour l'autre anticorps monoclonal de lapin contre le RUNX1 que nous avons dans le catalogue, le ab133261. Cet anticorps est testé en Western blot et sur coupe paraffine. Our lab has experienced some issues with item 92336, please see the comment below:
We used the antibody Anti-RUNX1 ab 92336 that we ordered from you a few weeks ago in parallel with Anti-RUNX1 ab23980, that another laboratory gave to us, on glyoblastoma cells as positive control, and ab23980 worked much better. Since we'll have to work with hematopoietic cells that have surely a positivity less extensive than glyoblastoma, is it possible for us to receive ab23980 as substitution of ab 92336?

I appreciate the time you have spent in the laboratory and understand your concerns. It is regrettable the results have not been succesful.

I purchased a vial of ab92336 after receiving assurances that it was specific for Runx1.  I am afraid I will need to get the purchase order number on Monday when the office staff are available but I attach for your attention a presentation to show that this antibody also reacts with Runx3 with good affinity.  We work on murine lymphomas which overexpress the Runx proteins.  On the blot I have included 4 that express high levels of Runx3, Runx1 &3, Runx3 and Runx2 (from left to right) through retroviral insertion.  The top blot is probed against a mouse monoclonal antibody that recognizes all three Runx proteins through their shared RUNT homology domain.  The lower blot is the same membrane stripped and reprobed against Ab92336. As you can see the best reactivity is against the Runx3 proteins.  On the second slide I have included a confocal micrograph of murine lymphomas.  The first two express Runx1 and Runx3 while the RHS panel is from a lymphoma that only expresses Runx3.  Again the abCam antibody is picking up Runx3 very effectively.  I would appreciate your comments but I would also like my money back since we require an antibody that specifically recognizes Runx1 and I was assured that this would be the case with Ab92336. I look forward to hearing from you.
