Target

  • Relevance

Function: Major acute phase reactant. Apolipoprotein of the HDL complex. Tissue specificity: Expressed by the liver; secreted in plasma. Disease: Note=Reactive, secondary amyloidosis is characterized by the extracellular accumulation in various tissues of the SAA protein. These deposits are highly insoluble and resistant to proteolysis; they disrupt tissue structure and compromise function. Note=Elevated serum SAA protein levels may be associated with lung cancer. Similarity: Belongs to the SAA family. PTM: This protein is the precursor of amyloid protein A, which is formed by the removal of approximately 24 residues from the C-terminal end.

    Answer

    Thank you for your email.

Finally, I have checked the sequence homology between human and horse SAA proteins, the homology is quite low with matching stretches.

    http://www.uniprot.org/blast/uniprot/2012101621EV411P6G?alignment=27

P0DJI9 is for human SSA protein and F6PJF6 is the ID for horse protein.

In order to further check whether the antibodies ab687 and ab18711 would be suitable, I have contacted the developer laboratory of these antibodies and asked if they can match the epitope of these antibodies with horse protein sequence. I will send you the details soon.

The other antibodies we have in catalogue are mostly against the full length human proteins so I am sorry we may not be able to support the horse species cross reactivity. We however have rabbit monoclonal that have >95% immunogen sequence homology with horse so this antibody should cross react with horse.

    https://www.abcam.com/Serum-Amyloid-A-antibody-EPR4134-ab108992.html

Along with ab108992 you can search an anti SAA antibody which detects N-terminal end of protein, to use these antibodies in sELISA. I am sorry we do not have N-terminal specific products.

I hope this information will nevertheless helpful. Please do not hesitate to contact me with further inquiries.

I am interested in purchasing two monoclonal anti-serum amyloid A antibodies for the purpose of developing an assay.
Can you tell me if the following antibodies are compatible pairs? ie. will steric hindrance occur on the target SAA molecule if both antibodies are used in an assay?Ab687
    Ab187313
Alternatively, can you recommend a known compatible anti-SAA pair?
I would also like to enquire about long-term availability of your products and up-scaling for manufacturing.

    Question

Phone call requesting information on if anti-Serum Amyloid A antibodies ab687 and ab81483 are likely to work in native western blotting.

    Asked on Mar 23 2012

    Answer

    Thank you for contacting us.

I have looked into what applications anti-Serum Amyloid A antibodies ab687 and ab81483 have been used for in order to ascertain if they are likely to detect the native epitope of Serum Amyloid A in order to perform native western blotting.

Both ab687 and ab81483 have been used in ELISA where the native epitope is presented. The ELISA carried out with ab687 was performed with the endogenously expressed protein, whilst the ELISA performed with ab81483 used recombinantly expressed protein. Both of these proteins are therefore likely to also be able to detect the protein when performing a native western blot. However, as we have not carried out this experiment I cannot guarantee that it will.

I also looked into what protocol had been used in performing immunoprecipitation with ab81483, however, the lab were unfortunately able to confirm that they had not tested the antibody in this application and that the informationhad been added to the datasheet in error. The datasheet has now been updated to reflect this.

I hope this information has been of help. If you require any further information please do not hesitate to contact us again.
