NAD-dependent protein deacetylase. Activates or deactivates mitochondrial target proteins by deacetylating key lysine residues. Known targets include ACSS1, IDH, GDH, SOD2, PDHA1, LCAD, SDHA and the ATP synthase subunit ATP5O. Contributes to the regulation of the cellular energy metabolism. Important for regulating tissue-specific ATP levels. -

SIRT3 was immunoprecipitated from 0.35 mg mouse liver lysate with ab246522 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab246522 at a 1/1000 dilution (0.45 μg/ml). VeriBlot for IP Detection Reagent (HRP)(ab131366) was used at 1/5000 dilution.
Lane 1: Mouse liver lysate 10μg
Lane 2: ab246522 IP in mouse liver lysate
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab246522 in mouse liver lysate.Blocking and dilution buffer and concentration: 5% NFDM/TBST.
