Target

  • Function

Receptor for somatostatins-14 and -28. This receptor is coupled via pertussis toxin sensitive G proteins to inhibition of adenylyl cyclase. In addition it stimulates phosphotyrosine phosphatase and PLC via pertussis toxin insensitive as well as sensitive G proteins. In RIN-5F cells, this receptor inhibits calcium entry by suppressing voltage dependent calcium-channels.
  • Tissue specificity

Cerebrum and kidney. In lesser amounts in jejunum, colon and liver.
  • Sequence similarities

Belongs to the G-protein coupled receptor 1 family.
  • Cellular localization

    Cell membrane.

Question

he data sheet indicates that the predicted band size for this antibody is 41 kDa, yet on the western-blot image for this antibody (on Abcam website) a band of 75 Kda is shown. Is it a mistake? Thank you for clarifying.

Asked on Jul 15 2015

Answer

The unprocessed Somatostatin Receptor 2 is about 41kDa, and then its molecular weight increases to ˜80 kDa at maturity.

The specificity of the ab134152 was also examined in a sstr2-deficient mouse model. Please refer to the two publications attached (contact Scientific Support). They should help further clarify.

Question

Does this antibody detect a special variant of SSTR2 (SSTR2a or SSTR2b), or does it detect all of the variants.

Asked on Nov 20 2013

Answer

This antibody, 3582-1/ab134152, reacts with an immunogen located at the C terminal residues of the canonical form of SSTR2. There is an alternate form of SSTR2 that expresses a different C-terminal sequence. Based on information in Uniprot, http://www.uniprot.org/uniprot/P30874#section_seq, it seems this antibody will not detect 'Isoform B' as defined on the webpage.

Inquiry: My question concerns the Anti-Somatostatin Receptor 2 antibody UMB1. Does it bind to both sst2 splice variants sst2A and sst2B? Or just to sst2A? Looking forward to hearing from you.

Asked on Nov 15 2013

Answer

I can confirm that there is no immunogenic homology with the SSTR2B protein so this antibody should bind to SSTR2A form only.
