General notes
This antibody was developed as part of a collaboration between Abcam and the lab of Dr. Kevan Shokat at University of California at San Francisco.
This antibody is useful for studying kinase-substrate relationships of interest. The antibody is also context independent in that it can recognize serine, threonine, and tyrosine thiophosphorylated residues in a multitude of kinase phosphorylation consensus motifs.
In order for this antibody to work in WB and IP, the customer needs to perform a specific kinase reaction using ATP gamma S and PNBM.

ab133473, at 1:5000, immunoblotting immunoprecipitated samples pre-resolved by SDS-PAGE. Myelin basic protein (MBP) was thiophosphorylated by AS GSK3 beta (GSK3 beta also catalyzes an auto thiophosporylation reaction). Samples were treated with or without p-nitrobenzyl mesylate (PNBM) for 1 hour at room temperature. PNBM was removed by size exclusion and 1mg/ml of HeLa cell lysate was added to samples.
ab133473 was pre-incubated with protein G beads and was added to sample for an immunoprecipitation at 4°C overnight:
(1) Thiophosphorylated MBP
(2) PNBM treated thiophosphorylated MBP sample
(3) HeLa cell lysate
(4) HeLa cell lysate with PNBM treated thiophosphorylated MBP sample
