      ab112997 at 1ug/ml staining Transketolase in HeLa cells by Immunocytochemistry (4% paraformaldehyde fixed and 0.1% Triton X-100 permeabilized) followed by Alexa Fluor® 594 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1 hour (red). Nuclei were stained with DAPI (blue). Non specific fluorescence was reduced using a signal enhancer.

      IHC image of Transketolase staining in Human normal colon formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab112997, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

      For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

    I have been using one of your anti-transketolase antibodies (ab112997) with good success. I have one question regarding the specificity. Is this particular antibody specific for transketolase (TKT) or does it also recognize the other transketolase family members, TKTL-1 and TKTL-2? The product data sheets that accompanied the purchased antibody didn't say and I couldn't find it on the website, either.

    Thank you for contacting Abcam regarding ab112997.


    Our TKT antibody was developed from a shotgun project, thus we do not exactly know the epitope of the antibody. The antibody was validated with Hela cells which express both TKT and TKTL1. IP resulted a single band and confirmed as TKT. No trace of TKTL1 was detected. The sequence similarity between TKT and TKTL1 or TKTL2 is 68% and 63% respectively. I can't ensure you for 100% since we did not test other tissues, but the evidence so far shows this antibody is specific to TKT.

    Thank you for your inquiry. We advise our customers to store this antibody at 4C. We do have data that storage at 4C will not affect the quality of this antibody. It might well be possible to freeze this antibody without experiencing quality loss and I would suggest to aliquot it to avoid freeze/thaw cycles in this case. But this has not been experimentally tested by us and it is not covered by our Abpromise guarantee. I strongly recommend to store ab112997 as stated on the datasheet to make sure it the Abpromise still applies.
