Relevance
The Vav family are Rho/Rac guanosine nucleotide exchange factors (GEFs), consisting of three members in mammalian cells (Vav, Vav2, Vav3) and one in nematodes (CelVav). First discovered based on its transforming properties, Vav is expressed mainly in hematopoietic cells and a few non-hematopoietic tissues, such as the pancreas and tooth enamels. As a signalling transducer, Vav is involved in T-cell activated transduction of T-cell antigen receptor (TCR). T-cell stimulated and tyrosine phosphorylated Vav acts as a catalyst in the exchange of guanosine nucleotides on Rac-1, a GTP binding protein. Using a mouse model, Vav expression has been determined to play an essential role in the cyctosketetal, proliferative, and apoptotic pathways for developing lymphoid cells and its signal response. -

Anti-Vav proteins antibody [EP482Y] (ab40875) at 1/2000 dilution + Jurkat cell lysate at 10 µg

Overlay histogram showing Jurkat cells stained with ab40875 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab40875, 1/100 dilution) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-rabbit IgG (H+L) (ab96899) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (1µg/1x106 cells) used under the same conditions. Unlabelled sample (blue line). Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter.
